Abstract of PhD thesis:
Searching new antibiotics has increased worldwide because of the serious problem of antibiotic
resistance among the microbes. The recent discovery of novel primary and secondary metabolites
from taxonomically unique population of actinomycetes suggest that these organisms could add a
new dimension to microbial natural product research. The history of new drug discovery processes
shows that novel skeletons have, in the majority of cases, come from natural sources. Today, the
emphasis is on the exploration of unusual and previously ignored ecosystems. Actinomycetes from
several unexplored environments have been intensively studied in last few decades for novel and
potent molecules. In this scenario, this study was focused on solitary wasp mud nest, swallow bird
mud nest, earthworm castings, hot water springs, and plant endophytes as well as soil samples
collected from different locations of Uttarakhand other than unusual habitats. On the basis of the
evidence obtained from the present study some general conclusions can be drawn that the solitary
wasp mud nest, swallow bird mud nest, and earth worm castings are very productive ecosystems with
regard to the occurrence of microbial flora. The results of the present study strongly support the idea
that actinomycetes species isolated from solitary wasp mud nest, swallow bird mud nest and
earthworm castings possess a significant capacity to produce compounds having unique antibacterial
activity as compared to terrestrial isolates. The isolates 8(1)*, MN 9(V), MN 2(6) and EWC 7(2) were
found to be most prominent in the terms of desirable activities. These isolates were found to have
excellent antimicrobial potential against several drug resistant bacterial pathogens. Preliminary
characterization of the metabolites produced by Streptomyces sp. 8(1)* and Streptomyces sp. EWC
7(2) indicates that it may be novel metabolites however further studies are required to elucidate its
structure to confirm novelty. The activity of gentamicin and cefotaxime were enhanced by the
presence of the metabolite produced by Streptomyces sp 7(2) against Gram-positive and Gram
negative bacteria including drug resistant strains. Similarly, the activity of piperacillin, gentamicin and
penicillin showed significantly improved activity against both Gram-positive and Gram-negative
bacterial strains when combined with the metabolite of Streptomyces sp. 8 (1)*. However, further
elaborate chemical, biochemical and pharmacological studies are necessary to develop these active
principles to the level of a drug.
