AP Biology Enzyme Lab Questions 1. 2. 3. 4. 5. 6. 7. 8. 9. Pre-Lab Questions In this lab you will be testing the factors that influence enzyme activity. What is the substrate you will be working with? What is the enzyme you will be working with? From which organism will it be obtained? What will be the byproducts of the observed reaction? How will we be measuring the rate of the reaction in our lab? Parts A &B will be completed by all groups. Below, draw & label the experimental apparatus you will be using in the lab. What will first be put into the glass reaction chamber? What will be pipetted into the reaction chamber after? Why does the reaction chamber need to immediately be stoppered and placed into the water bath? What will happen if you take too long, or if the stopper isn’t fully sealed in the chamber? How should you prepare the setup of the lab to make sure you accurately obtain all of the bubbles formed in the reaction chamber? Please explain which factors you could be testing if you are given labs C, D, E, or F to complete. Give specific details. Post Lab Questions: 1. Don’t forget to complete the graphs for Parts A & B! 2. Calculate the rate of reaction (slope) for each part of the experiment you completed. The formula is: m= y2-y1 x2-x1 3. Summarize the effect of enzyme concentration on enzyme activity. Explain the mechanisms that cause this effect. 4. What would cause the rate of enzyme activity to level off on the enzyme concentration graph? 5. Draw an “idealized” graph to illustrate the relationship between enzyme concentration and rate of enzyme activity. 6. After an enzyme is mixed with its substrate, the amount of product formed is determined at 10second intervals for 1 minute. Data from this experiment is show below. Time (sec) Product formed (mg) 0 0.00 10 0.25 20 0.50 30 0.70 40 0.80 50 085 60 .85 a. What is the initial rate of this enzymatic reaction? Show work. b. What is the rate at 50 seconds? Show work. c. Why is the rate at 50 seconds different from the initial rate? 7. Now complete the questions that correlate with the lettered lab you completed (C, D, E, or F)
