Supplemental Figure 2. #1 #4 #2 #3 #5 A: Oligomycin [2 µM] (inhibitor of ATP synthetase, to shut down OXPHS); B: FCCP [0.5 µM] (uncoupler, dissipates membrane potential, providing unlimited protons for ATP synthesis); C: Rotenone [5 µM] (inhibitor of Complex I, blocking ETS) #1: Basal Respiration Capacity #2: ATP turnover #3: Proton leak #4: Max. Respiratory Capacity #5: ROS (non-mitochondria related) Representative mouse colon crypt bioenergetic profile from the Seahorse XF24 Analyzer. Fresh isolated crypts from mouse colon were subsequently challenged with an ATP synthetase inhibitor (Oligomycin), respiratory chain uncoupler (FCCP), and complex I inhibitor (Rotenone). The profile was recorded as oxygen consumption rate (OCR) throughout the analysis. Based on the different OCR response to specific electronic transfer chain inhibitors, basal respiration capacity, ATP turn over, proton leak, maximum respiratory capacity, and ROS were calculated as indicated.
