3_ Bio-chemical tests (physiological)
Bio-chemical tests:
It is the most important studies of microorganisms in order to identify them is the
study of physiological or biochemical traits, which appear as a result of doing many
of the enzymes
Usually bacteria differed in how they contain these enzymes and this leads to the differences
we see from
the results of the tests of bio-chemical.
Enzyme hydrolysis test:
Different bacteria in the analysis of organic substances with large molecules such as starch
and cellulose, gelatin etc. and analyzes in the presence of water into smaller particles so
easily absorbed cells
Starch hydrolysis :
For this test of great importance to identify the different bacterial species. 
Bacteria that have the ability to analyze the starch produces four types of enzymes: 
Alpha amylase
)a
Beta amylase )b
Amelo 1-6 glococedase
)c
maltase )d
Name of Experiment:
Starch hydrolysis
The objective :
to determine if the bacteria can hydrolysis starch
incubate all plates in an inverted position for 24 to 48 hours at 37
analyze cellulose:
Some bacteria analyze cellulose either under aerobic conditions or anaerobic .
There are two theories for the analysis of cellulose
Theory 2-enzymes :
cellulose
cellabiose
Glucose
Theory of multiple enzymes:
Cellulose
cellabiose
polyanhydro glucose
glucose

Name of Experiment:
analyze cellulose (aerobic and anaerobic) .
The objective :
to determine if the bacteria can analyze cellulose (aerobic and anaerobic)
method :
1-analyze cellulose aerobic.
2-analyze cellulose anaerobic .
