Table S1 List of MS-identified O-GlcNAc metabolically labeled proteins and their assigned peptide counts
derived from proteomics with probe Biotin-Diazo-Alkyne.
O-GlcNAc metabolically labeled cell lysates were clicked to Biotin-Diazo-Alkyne. After enrichment on streptavidin
beads, the bound proteins were eluted by boiling for 5min. Eluted proteins were separated on SDS-PAGE. The
protein bands were then sliced from the gel and subjected to in-gel trypsin digestion for MS analysis. Proteins that
were identified in both probe Biotin-Diazo-Alkyne and dbOGAP database were in red font. Proteins that were
identified in both probe Biotin-Diazo-Alkyne and probe Biotin-DIBO-Alkyne were highlighted in yellow color.

derived from proteomics with probe Biotin-DIBO-Alkyne.
O-GlcNAc metabolically labeled cell lysates were clicked to Biotin-DIBO-Alkyne. After enrichment on streptavidin
beads, the bound proteins were eluted by boiling for 5min. Eluted proteins were separated on SDS-PAGE. The
protein bands were then sliced from the gel and subjected to in-gel trypsin digestion for MS analysis. Proteins that
were identified in both probe Biotin-DIBO-Alkyne and dbOGAP database were in green font. Proteins that were
identified in both probe Biotin-Diazo-Alkyne and probe Biotin-DIBO-Alkyne were highlighted in yellow color.
