Interpretation of any positive or negative staining must be complemented with the evaluation of proper controls and must be made within the context of the patient’s clinical history and other diagnostic tests. A qualified pathologist must perform evaluation of the test. Summary and Explanation: Ribonucleoside-diphosphatereductase large subunit (RRM1) is one of two non-identical subunits that constitute ribonucleoside-diphosphatereductase, an enzyme essential for the production of deoxyribonucleotides prior to DNA synthesis in S phase of dividing cells. Studies have shown that RRM1 controls cell proliferation through deoxynucleotide production and metastatic propensity through PTEN induction. RRM1 expression is significantly correlated with the expression of ERCC1 and PTEN in non-small-cell lung cancer (NSCLC). Tumors with high expression of RRM1 showed slow progression. Staining Recommendations: Antigen Retrieval Solution: Use Tris EDTABuffer(PathnSituCat # PS009) as antigenretrieval solution Heat Retrieval Method: Retrieve sections under steam pressure for 15 min using PathnSitu’s MERS (Multi Epitope Retrieval System) then allow solution to cool for 10 minutes then transfer tissue sections/slides to distilled water. Primary Antibody: Cover the tissue sections with primary antibody and incubate for 30 min at room temperature when used PathnSituPolyExcel Detection System. Detection System: Refer to PathnSituPolyExcel detection system protocol or manufacturer’s detection kit staining protocol when used other vendor detection system.
