A gene is considered to be down-regulated if the ratio of medians is less than 0.7. In a DNA microarray study
testing normal human epidermal keratinocytes, 0.1% TRIglyphix® Sense was shown to down-regulate the
expression of IL-1β and IL-1RAcP. As a result, this data indicates TRIglyphix® Sense may help decrease the
production of pro-inflammatory cytokines while also reducing the inflammatory response generated through
the Interleukin 1 receptor.

In the study below, MatTek full thickness tissue models were used to measure a reduction in IL-1α and PGE2. 1%
Sodium Dodecyl Sulfate (SDS) was used to trigger the inflammatory response. TRIglyphix® Sense was shown to
reduce the expression of IL-α and PGE2 by as much as 41% and 49%, respectively.* It was also shown to improve cell
viability compared to cells treated with SDS alone.

Objective:
To evaluate the performance of TRIglyphix® Sense to reduce redness
and irritation from shaving over a period of seven days.

Panelists with similar average irritation were divided into three
groups of five people. Panelists were to shave every day for seven
days and immediately apply the treatment to the shaved area.
Observations were made at Day 0 (baseline), Day 3 and Day 7. A
clinician then evaluated the severity of folliculitis (0-10 scale, best
to worst). In addition, photogrammetrix grading analysis software
was used on a panelist from each group to show changes in the
three dimensional profile of hue, value and chroma.
Results:
TRIglyphix® Sense proved to be soothing and effective for all
users involved in the study. The clinician’s results showed an
average decrease in redness by as much as 70.59% when using
TRIglyphix® Sense. Photogrammetrix results also showed
an average decrease in redness by as much 89.91%. The two
panelists, (Figure 3 & 4), illustrate the clinician’s findings that using
TRIglyphix® Sense will help to visibly reduce redness and razor
burn. This supports the use of TRIglyphix® Sense as an effective
soothing ingredient for sensitive skin.

To measure the soothing benefits TRIglyphix® Sense can deliver
by reducing sting discomfort following application of lactic acid.
• 10 panelists (2 male, 8 female) ages 21-50

A clinician applied 10% lactic acid (aqueous solution) to both
sides of panelists face (nasolabial fold and upper cheek areas) to
induce a feeling of discomfort. After five minutes, the clinician
then applied the placebo to one side of the face and the placebo
with 2% TRIglyphix® Sense to the other side in a blinded fashion.
Following 2.5 and 5.0 minutes after application, subjects rated
discomfort/sting perception on a scale from 0 (no discomfort) to
3.0 (severe discomfort) in increments of 0.5.
Results:
TRIglyphix® Sense was shown to help soothe discomfort by 59%
after five minutes, when compared to the placebo. The results
support the use of TRIglyphix® Sense in cosmetic and personal
care formulations to minimize the potential aggravation caused by
irritating cosmetic ingredients.
To determine if TRIglyphix® Sense could deliver an additional
soothing benefit to formulations that are developed specifically for
sensitive skin, (e.g. free of fragrances, dyes and actives.)
• 10 male panelists, ages 18-49

o Facial Cream with 2% TRIglyphix® Sense
A clinician applied the placebo to one side of the panelists face
and the cream with 2% TRIglyphix® Sense to the other side
(similar to Study 1). Following 2.5 and 5.0 minutes after application,
subjects rated discomfort/sting perception on a scale from 0
(no discomfort) to 3.0 (severe discomfort) in increments of 0.5.
Results:
TRIglyphix® Sense was shown to help soothe burning and
stinging sensations by 73% after five minutes, when compared
to the placebo. This suggests TRIglyphix® Sense offers excellent
soothing properties that can help make a mild formula even
gentler to the skin.

Sensitive skin is extremely susceptible to damage, specifically to harsh environmental conditions and chemical insult.
It is often prone to drying and cracking. When damage occurs, steps must be taken to prevent instances of acute
inflammation from becoming a chronic issue. Important factors for optimizing the skin’s repair include fibroblast
proliferation, collagen, elastin, hyaluronic acid synthesis and MMP-1 inhibition.
