Poly(A) RNA was enriched from replicate 100 µg samples of total RNA using MicroPoly(A)Purist, Poly(A)Purist MAG, and four other commercially available poly(A) RNA selection kits. One quarter of each recovered mRNA sample was analyzed by formaldehyde agarose gel electrophoresis. The RNA in the formaldehyde agarose gel shown in Figure 1 was transferred to a nylon membrane and hybridized with 32P-labeled RNA probes for β-actin, GAPDH, and cyclophilin (respective transcript sizes are 2.1 kb, 1.5 kb, 0.7 kb). A comparison of the signals obtained for each sample demonstrates that the mRNA isolated with the Poly(A)Purist Kits yield more mRNA than do competitors’ kits. Identical total RNA samples were processed with each kit and careful evaluation of the subsequent mRNA revealed that the two Ambion kits provided the lowest level of residual rRNA contamination. Ethidium bromide staining on a denaturing agarose gel and Northern analysis confirmed the observations. These Poly(A)Purist kits are available in two formats—one uses oligo(dT) cellulose-based selection: Poly(A)Purist and MicroPoly(A)Purist, and one that utilizes oligo(dT) magnetic bead-based purification: Poly(A)Purist MAG kit.
