Facility and Resources-Stem Cell Core Facility (SCCF) General Introduction The Stem Cell Core Facility (SCCF) at The Johns Hopkins Institute for Cell Engineering (ICE) was opened September 2011 to help human stem cell researchers. The missions of SCCF is to maintain stem cell lines (hES/hiPS), to provide researchers with stem cell lines and related materials, to perform validation and quality-control of stem cell lines, to provide the technology for stem cell derivation, culture, differentiation and genetic modification, and to train and assist end users with stem cell related techniques. The facility will provide the community with services ranging from simple hESC maintenance to advanced techniques on a fee per service basis. Personnel Director: Valina L. Dawson, Ph.D. Co-Director: Hong-jun Song, Ph.D. Manager: Hyesoo Kim, Ph.D. Research technologist: Shravya Kommineni, M.S. Laboratory space The Laboratory is located on 7th floor of Edward Miller Research Building. It consists of 1000 sq ft of space and contains a tissue culture room, bench workspace and an office. In addition, a centrifugation and storage room, microscopy room and a walk-in cold room are available as common space. Security access is in place for the TC room. Equipment Four class II biosafety cabinets including one for virus work, eight CO2 incubators, one IVF station with two attached stereoscopies and a monitoring system, one centrifuge and two inverted microscopies are housed in the tissue culture room. A PCR machine and electrophoresis system are available for RT-PCR and two rockers for immunostaining are housed in the non-TC room. For storage of biological samples, we have one -80°C deep freezer, one liquid nitrogen freezer which maintains up to 20,000 frozen vials, two 4°C refrigerators and one -20°C freezer. Flow cytometry and gel doc system apparatus are also available as a common resource of ICE. Services Providing different types of cells Custom hiPSC generation Differentiation of hES/hiPS cells Genetic modification of hES/hiPS cells Individual and group training courses Other custom experiments Others Human cells and viruses are registered annually with the JHU Biosafety Office and are handled under authorization of the Johns Hopkins Institutional Stem Cell Research Oversight (ISCRO).
