Flow Check® Flow Cytometry Particles & Sets
INTRODUCTION
The first flow cytometers became commercially available in the early
1970s. Since then the use of flow cytometry has expanded dramatically and
become an invaluable tool for clinical diagnostics, cancer biology research,
cell biology, and many other fields that examine cellular and subcellular
components. To a large degree, the increasing popularity of flow cytometry
is due to the fact that it allows large numbers of biological or non-biological
particles to be rapidly analyzed and quantitated. In addition, because of the
ability to examine multiple signals at one time, flow cytometric analysis allows
the simultaneous examination of several parameters in a single sample.
Flow cytometers are complex instruments that require the fluidic system,
excitation optics, and detection optics to be in optimal working order. The
precise alignment of these systems insures the highest levels of sensitivity
and repeatability. In addition to
optimizing the overall performance
of the flow cytometer, it is
also necessary to set up the
instrument for the particular
type of samples being analyzed.

CHARACTERSTICS OF FLOW CHECK® FLOW CYTOMETRY
PARTICLES
Size and Uniformity Flow Check® Flow Cytometry particles are ~6µm in
diameter and manufactured on site to strict standards of size uniformity.

The graph shows size distribution data on a typical lot of polystyrene
microspheres used to manufacture
Flow Check® Flow Cytometry
particles. The measured diameter
of the microsphere is 5.8630 µm
(volume analysis) with a standard
deviation of 0.2840 µm. Data were
obtained using a Brookhaven
BI-DCP Disc Centrifuge.
Fluorescence Flow Check® Flow Cytometry particles are available in a
variety of fluorescent colors and intensities for calibrating flow cytometers.
Flow cytometric and spectroscopic analysis ensure consistent product
performance.

Flow Check® Intensity Calibration Sets Format Flow Check® Flow
Cytometry particles are ~6µm polystyrene microspheres in aqueous
suspension (1x PBS, 0.1mM EDTA, 0.1% BSA, 0.1% NaN3). They are
supplied in 5ml dropper bottles at a concentration of 1 x 107/ml. Flow
Check® Intensity Calibration Standards can be purchased as a set or
individually. Sets include an undyed particle and microspheres with four
different fluorescence intensities.

HANDLING AND STORAGE
Storage Store Flow Check® Flow Cytometry particle suspensions at 4˚C
when not in use. Prior to dispensing, particles should be well mixed by
completely inverting the bottle several times. For use, dilute one drop of the
particle suspension in 1ml of sheath fluid and mix well. Diluted particles
should be stored at 4-8˚C and protected from light. Diluted particles are
stable for 8-10 hours when stored in the dark at 4-8˚C.
Safety This particle suspension contains sodium azide. Sodium azide
may react with lead and copper plumbing to form explosive metal azides.
