PAA – Cell Culture and More
PAA is specialized in the manufacture and worldwide distribution
of cell culture products for research, development, diagnostic and
biopharmaceutical production. For more than 20 years we
accompany our customers in their daily laboratory routine and
provide them with high quality animal and human sera, media,
reagents and buffer solutions. Our products are manufactured in
three facilities based in Austria, Australia and Canada.
In parallel with our investment in facilities and capabilities we
have also committed considerable funds into new product development to satisfy the requests of our research based customers.
We have expanded our product range with new and innovative
media and several new reagents.
Beside the excellent product quality, our main aim is to provide
outstanding customer service. In consequence of this we expanded our product portfolio with disposables such as plastic labware
to enable our customers to purchase their laboratory equipment
conveniently from one source.
The plastic labware is designed by leading manufacturers, using
high precision injection molding, polymer technology and surface
chemistry. Quality characteristics such as the absence of endo­
toxins, non-cytotoxicity as well as exceptional optical clearance
and good cell performance are the basic principles of our cell
culture products.
For the area of cell imaging we provide highly innovative products
for cell based assays, fluorescence microscopy and high content
screening. They feature high plate planarity, low background
interference and high light transmission for challenging experiments.
Our company is known for its focus on high throughput laboratories as well as the support of smaller research groups. Therefore,
we offer our new product line in different purchase units to
satisfy the individual needs of our customers.
Quality Control
The basic requirement for the reproducibility and comparability
for cell and tissue culture in the field of in vitro research is ­first
class quality standard.
Sterility
Sterilization is a process in which surfaces of materials and
objects are freed from living organisms. As a 100 % security can
not be guaranteed, a reduction is required, so that a SAL-value
(Sterility Assurance Level) of 10-6 is reached. This means that in
a million equally treated units of the sterilized products only one
augmentable micro-organism should be included. This SALvalue can be achieved by gamma irradiation, beta irradiation
and ethylene oxide sterilization.
RNA/RNases and DNA/DNases
DNases and RNases belong to a group of enzymes that destroy
nucleic acids. They protect living organisms against the invasion
of foreign DNA and foreign RNA by hydrolyzing the phosphodi­
ester bond between the 5'-phosphate group of a nucleotide and
the 3'-hydroxyl group nearby. DNA/DNases and RNA/RNases have
a great potential for transmission so that protection during working processes is necessary.
Pyrogens
Pyrogens are high molecular weight substances of polymerous
structure which might effect cell growth and proliferation. They
can either be an endotoxin or an exotoxin. Endotoxins are natural
compounds of the cell wall of gram-negative bacteria and potentially toxic. The biological activity of endotoxin is associated with
the lipopolysaccharides (LPS) of the bacterial membrane.
As pyrogens are heat stable and cannot be destroyed by gamma
irradiation it is necessary to test for their absence. The Limulus
Amoebocyte Lysate Assay (LAL assay) is a very sensitive method
for the detection of the presence and quantity of bacterial endotoxins. The LAL test complies with the FDA guidelines.
Cytotoxicity
Cytotoxicity describes the ability of distinct chemical substances
to damage cells or tissues. Especially for reliable toxicological
studies in cell culture research the quality of culture vessels is of
high relevance. It must be guaranteed that they are free of cytotoxic substances to prevent false positive results. The cytotoxicity
is determined by using mammalian cells according to EN ISO
10993.
Materials
Most of our plastic labware is made of either polystyrene or polypropylene. Polystyrene (PS) is a high quality polymer with an
excellent optical clarity. This feature facilitates the optical control
of cells cultivated in PS flasks, wells and dishes. PS is also applicable for the manu­facturing of serological pipettes and tubes where
precise mea-surements are necessary.
Polypropylene (PP) is a linear hydrocarbon polymer used for many
different applications. PP is characterized by a high chemical resis­
tance to many chemical solvents, bases and acids. It shows thermal stability in a temperature range of −80 °C to +121 °C and is
therefore autoclavable. Due to its high mechanical resistance PP
is used for products which are exposed to pressure occurring e.g.
at high speed centrifugation.
Surface Treatment
Our plastic labware used for the cultivation of adherent cells is
plasma treated. This surface technology is based on the use of an
ionized gas at a certain pressure to increase the surface energy.
As a result the polystyrene surface of the cell culture vessels is
more hydrophilic and allows optimal cell attachment.

Cell Culture
In research cell culture flasks are routinely used for the cultivation of eukaryotic cells. They are first class products for adherent
cells as well as for suspension cultures and applicable for different requirements.
PAA offers standard and filter cap cell culture flasks of excellent
optical clarity. The filter caps are equipped with a high quality
membrane with a pore size of 0.22 µm allowing consistent gas
exchange. Cell culture flasks with green caps are suitable for
adherent cells, providing a surface treated zone for optimal cell
adhesion and growth. Flasks with white caps meet the requirements of suspension cells.
The design of the flasks provides flexibility in media volume and
saves incubator space. In addition, it facilitates the accessibility
with cell scrapers and serological pipettes in an optimal way. The
cell culture flasks are easy to stack and stand firmly in the incubator. The angled neck of the high profile flask reduces the risk
of wetting the inside of the cap with medium and thus provides
additional protection against contamination.

Immunoplates
Cell culture dishes are applicable as alternative to cell culture
flasks and appropriate for different requirements.
PAA offers cell culture dishes of excellent optical clarity. High
quality surface treatment of the growth area enables constant
results. Easy stackability ensures a firm stand in the incubator.
A raised ring around the circumference of the dish provides a
secure grip of dish and lid. This reduces the risk of contamination due to unintentional removal of the lid. Cams inside the lid
allow constant ventilation in a closed system.

Cell scrapers and lifters allow an efficient and gentle removal of
adherent cells from the cultivation surface. They can be used as
an alternative or in addition to enzymatic detachment solutions.
The cell scrapers are designed for optimal contact to the culture
vessel and appropriate to flasks, dishes and plates. Even in culture
containers with poorly accessible surfaces the free rotatability of
the blade enables easy and complete scraping of attached cells.
The thin and flexible blades of the scrapers are made of polyethylene, a material that does not affect the cells.
The cell lifter is especially suitable for gentle harvesting of cells
and tissues from dishes and plates. The one piece design with a
small and a big blade on one handle facilitates working with
diverse culture vessels.

Multiwell tissue culture plates are used in all areas of cell culture
and molecular biology.
PAA provides multiwell plates in several sizes and for different
applications. All of them are cell culture tested and designed for
reduced cross contamination risk. The arrangement of each well
standing on its own and corresponding condensation rings on

Colorimetric assays, e.g. fluorescence or luminescence measurements are more and more common in cell culture research. To
avoid negative side effects such as background interference,
autofluorescence or cross talk the use of black or white pigmented microplates is recommended.
PAA provides two different high quality 96 well microplates,
suitable also for various high throughput assays. High signal-tonoise ratio with low background is a characteristic feature of
these plates.
The 96 well microplates are cell culture tested and designed for

Hemocytometers are used in daily routine in cell culture, microbiology and haematology. The preparation of traditional glass
chambers is a time-consuming and not reliably reproducible
process that often ends with broken cover slips.

Confocal Dishes
The in vitro cultivation of cells under laboratory conditions is
common practice for a huge variety of applications. The range
of cell types is almost limitless, e.g. plant and insect cells as well
as avian and mammalian cells.
PAA offers confocal dishes made of polystyrene with coverglass
bottom or removable coverglass insert. The dishes facilitate
preparation of cells for microscopic analyses. They are suitable
for use in high resolution and inverted microscopy, fluorescence
imaging in living cells, confocal microscopy, phase contrast
microscopy and micro-manipulation or anywhere where highest
image quality is desired. A raised ring around the circumference

Serological pipettes are an essential tool for precise volume
measurement. In contrast to glass, plastic pipettes provide the
advantage of constant sterility due to individual packaging.
PAA offers disposable serological pipettes that are single
wrapped in plastic/paper packaging with peel-off function for
aseptic pipette removal. The serological pipettes are also availa-
ble in bulk packages, both sterile and non-sterile. High grade
polystyrene provides maximum transparency. To avoid suction of
liquid into the pipetting device all pipettes are supplied with a
cotton plug. A color code, following international standards,
facilitates the identifi­cation of the pipette size. The slim pipette
tip and the small diameter allow outstanding accessibility.

The isolation of cells or subcellular particles is the first step for
diagnostics or investigations in cell functionality or gene expression. The enrichment of cells from complex biological mixtures
such as blood and other body fluids requires the use of special
separation media. The blood sample has to be poured carefully
over the separation medium in a time consumptive and
­laborious procedure.
To simplify this process PAA provides a centrifuge tube with an
incorporated porous barrier. This feature prevents a mixture of
the sample material with the separation medium and a recontamination of the enriched cell fraction with unwanted blood

Separation of lymphocytes – easily done with LSM 1077
For the separation of lymphocytes or other cells out of blood special separation
media are required. The media are used for the production of density gradients for
the separation of blood components, which sediment during centrifugation due to
gravity. PAA offers the ready to use separation solution LSM 1077 based on ficoll
with a density of 1.077 g/ml. Optimal osmotic conditions and a physiological pH level

For greater flexibility in diagnostics PAA offers strip immunoplates with maxi- or medibinding surface. One strip consists of
8 wells and 12 strips fit into one frame, thus forming a standard
96 well plate.
The wells of PAA38096X and PAA38196X are stabilized with a
polystyrene bar and thus fixed for easy and safe handling.
With PAA38296X and PAA38396X the individual wells can be
easily broken off separately. The number of performing tests can
thus be precisely adjusted to the number of samples. The frame
itself provides individual 96 wells for safe fixation of the wells.

PAA offers imaging dishes with a bottom made of cover glass
for imaging purposes which are highly demanding. The material
of this bottom is borosilicate glass and provides an excellent
planarity and focus control for imaging techniques with high
resolution. The 35 mm dishes are available with 145 µm or
175 µm glass thickness. For better microscopic orientation the
175 µm glass version is also offered with engraved micro grid.
Macroscopic orientation is supported by N-O-S-W marks. A grip
zone around the dish allows secure handling and enables an

Imaging Dishes CG
Cell Imaging
For convenient handling PAA offers Imaging Chambers that
allow culturing, staining and fixation of cells inside the same
vessel. They are available with cover glass (CG) or microscope
slides as bottom structure. Both types are available in 1 well,
2 well, 4 well and 8 well format. Imaging Chambers CG are
specially designed for high resolution live cell microscopy techniques. Imaging Chamber with microscope slides can be used
with long distance objectives.
PAA`s Imaging Chambers and lids are made of high performance polymers with better chemical resistance and temperature
stability than the commonly used polystyrene. The chambers are
sealed against the glass bottoms by biologically inert silicone
adhesive. At the end of the experiments the glasses can be
easily detached from the polymer chambers with minimal glue
residue.

CytoCapture products with square cavities. This type of geometry is especially designed for 3D guided cultivation of adherent
cells (e.g. epithelial cells). The upright growth of the cells allows
a more natural cell development with both apical and basolateral membrane sides. The microstructure assisted control of cell

Cryotubes are commonly used for storage of cells, sera, blood
components, bacteriological cultures and other fluids at low
temperature.
The cryotubes of PAA are made of clear high grade polypropylene. Using the same material for both vial and lid as well as the
exclusion of an O-ring reduce the risk of explosion during thawing to a minimum. The round bottom shape facilitates complete
transfer of the cell material to the cell culture flask or dish. The
starfoot base of the tubes enables easy handling within a working rack. The cryotubes are suitable for storage in refrigerators
or in the gas phase of liquid nitrogen as the material is stable at
temperatures down to -196 °C.
Excellent cooling and freezing conditions with PAA’s Cell Preservation Products
There are two common methods of cell preservation. Hypothermic preservation at temperatures above freezing allows short term storage of tissues and organs. Cryogenic
preservation at temperatures below freezing is used for the establishment of cell banks

reservoirs made of white polystyrene. The reservoirs are de-

The bottles are extremely lightweight, shatter-resistant and provide excellent gas barrier properties. The bottles are delivered in
brickpacks. Two or four brickpacks are double bagged and then
packed in master boxes.

In addition to the autoclave bags PAA offers a convenient
holder, which can be used on the bench and eases the handling
with contaminated biohazard material. This plastic coated
holder has three non-slip rubber feet and is suitable for bag size
200 x 300 mm.
Working with biohazardous materials requires correct
handling. In addition, the contaminated laboratory disposals like
cell culture flasks or pipette tips have to be destroyed in an
appropriate way before final removal.
PAA offers autoclave bags made of particular strong polypropylene film to use during daily laboratory routine. For autoclaving the bags should not be closed completely. They are autoclav-
