Exopolysaccharide (EPS) produced by a planktonic
microorganism through biotechnology. nocturshape™
blue ingredient takes advantage of the circadian
rhythms and targets nocturnin in adipocytes,
a protein involved in adipogenesis and lipid
accumulation. It reduces thigh contour and
smoothes skin relief, providing a general
improvement of cellulite appearance.

Properties
nocturshape™ blue ingredient provides a slimming
effect and smooths the skin relief due to a decrease
of fat nodules at night. It also helps to reinforce the
dermal matrix, enhancing the firmness of the skin.
The overall appearance of cellulite is improved.
Applications
nocturshape™ blue ingredient can be incorporated
into body care formulations with global anticellulite, slimming and firming properties, being
particularly active during the night.

Science
The most relevant biological cycles are circadian
rhythms, which comprise oscillations every 24
hours. The intracellular biological clock is controlled
by a network of feedback loops composed of
activators and repressors. The main clock genes
are CLOCK and BMAL1, regulating the expression
of hundreds of downstream circadian genes.
Among these genes, nocturnin shows a rhythmic
transcription pattern with a peak by early night
and it is greatly expressed in adipocytes. The
protein nocturnin promotes adipogenesis and fat
accumulation.
nocturshape™ blue ingredient is a biotechnological
active produced by a planktonic microorganism
(Halomonas sp.) isolated in Fuente de Piedra
Lagoon (Málaga, Spain). Its shallow waters, with a
high salt concentration, determines the presence
of abundant microorganisms and constitutes
a strategic point for migrating birds, especially
pink flamingos. This EPS reduces nocturnin
levels and thus lipid accumulation specifically at
nighttime. Besides providing a slimming effect,
it strengthens the dermal matrix through type I
collagen synthesis, helping to reduce visible fat
nodules characteristic of cellulite condition.

1. REDUCTION OF NOCTURNIN
PROTEIN LEVELS
Nocturnin levels (%)
Human subcutaneous preadipocytes were
synchronized to a day-night rhythm during
the differentiation. Cells were treated with
0.1 mg/mL nocturshape™ blue ingredient
concentrate for 6 h under induced night
conditions. Levels of nocturnin were
quantified by an ELISA.

2. INDUCTION OF LIPOLYSIS AT NIGHT
The same type of cells were synchronized again during the last days of differentiation and
treated with 0.1 or 1 mg/mL nocturshape™ blue ingredient concentrate. Glycerol release was
evaluated using a free glycerol assay kit. Differentiated cells were used as control.
3. DECREASE OF LIPID ACCUMULATION
Nile red staining was used for the quantification, by fluorescence, of lipids in human
subcutaneous preadipocytes, either non-treated or treated with nocturshape™ blue ingredient
concentrate.
