Pre-Filled Culture Vessel Modified Half Fraser Broth - Dehydrated For the Detection of Listeria spp. Culture Vessel The Speedy Breedy culture vessel is the disposable sterile unit that is required for each Speedy Breedy test. Speedy Breedy provides excellent culture conditions for rapid microbial growth. The culture vessel couples to the instrument so that internal pressure transients within (generated by microbial activity) can be measured by Speedy Breedy over the course of a programmed time and temperature culture protocol. Speedy Breedy can be used for a range of applications including contamination testing or microbial growth monitoring. Single use Speedy Breedy culture vessels are supplied sterile, either unfilled or filled with dehydrated culture medium. This Modified Half Fraser Broth filled vessel incorporates a ready to use culture medium in the form of encapsulated dehydrated powder that readily dissolves in an aqueous sample (c.a. 10 minutes). Each vessel contains sufficient medium for a working volume of 50 ml. This means that samples can be a full 50 ml volume or smaller samples can be used and topped up to 50 ml with sterile water making this product presentation highly versatile across a wide range of applications. General Culture Vessel Description The single use culture vessel totally encloses the microbial culture through the course of a timed test protocol. The vessel provides a suitable culture enclosure for media and organisms, and is sealed, separating it from the external environment, sensors and the operator, for safety and instrumentation protection. Gas usage or generation due to microbial respiration is contained so that linked pressure sensors in Speedy Breedy which houses and maintains the vessel during the test can monitor pressure events that indicate growth. The sealed vessel avoids contamination from external sources during a culture run. Features Speedy Breedy vessels manufactured from inert, impermeable plastic and rubber. Normal working range of 5OC below ambient to 45OC. High efficiency, non-invasive mixing system that performs three functions: 1. Homogenisation of essential nutrients and gases for optimal growth. 2. Rapid equilibration and accurate temperature maintenance. 3. Efficient gas transfer between headspace and culture medium for rapid conversion to detectable pressure change. Vessel pressure monitored indirectly by a sensor housed in the Speedy Breedy instrument. Protective barrier between the vessel contents, sensor and operator. Sample port with a removable cap including a self-sealing rubber septum with a tamper evident seal. Sample port allowing two distinct methods of inoculation, reagent addition or removal: 1. By removal of the cap allowing access to the vessel lumen via a 7 mm access port. 2. By piercing the cap septum with a hypodermic needle Modified Half Fraser Broth (Product Code BAC0029) Modified Demi-Fraser Broth is a selective medium which is used for the detection of Listeria spp. with the Speedy Breedy microbial respirometer, with applications in industries such as food production. A blend of peptone, tryptone, Lab Lemco powder and yeast extract provide a highly nutritious growth environment whilst sodium chloride maintains osmotic equilibrium and a phosphate buffer system controls pH. Nalidixic acid, acriflavine hydrochloride and a Bactest Selectivity Mix enhance the selectivity of the medium, improving growth of Listeria spp. organisms and suppressing Gram negative growth as well as the growth of non-Listeria spp. Gram positive organisms such as staphylococci and enterococci. The Modified Demi-Fraser Broth culture vessel should be used in conjunction with the Speedy Breedy Listeria test protocol to ensure optimum growth conditions and performance. A significant pressure event indicated by Speedy Breedy should be considered as presumptive for the presence of Listeria spp. In the event of any positive result, confirmation by appropriate culture, serological or molecular techniques is recommended. Precautions Following a positive result, Speedy Breedy culture vessels will contain a rich, live culture of Listeria spp. Speedy Breedy culture vessel design ensures operator safety. Culture vessel use, following Good Laboratory Practice, the recommended operating procedures and the recommended disposal procedures will ensure that operators are not exposed to infectious material. Disposal Used culture vessels are considered hazardous material and need to be treated as such. Health and Safety hazard assessments should be made prior to all microbiology work including waste disposal. For the purposes of disposal, culture vessel materials are compatible with standard procedures such as: Autoclave 121°C @15 psi for 15 minutes.
