Additional Instructions:
Service is currently offered only to tenants in Proteos on a trial basis.
Samples submitted must contain primer and DNA template topped up with water (if necessary) to 6 or 12µl
for 10 or 20µl reaction volume respectively.
Please kindly indicate ‘Require Cycle Sequencing’ in the remarks column upon submission online and
deposit the samples in the respective box as to avoid confusion with purified samples.
Cycle sequencing is only carried out overnight from Monday to Thursday and will be processed the next
working day. All reactions carried out are for normal plasmids (3-4kb insert) and PCR products (<4kb),
o
with annealing temperature at 52.5 C for 35 cycles. Please carry out your own cycle sequencing for
Genomic/BAC/YAC samples as these require conditions to be optimized.
For samples in 96-well plates, cycle sequencing is only carried out when there is a block available on one
of our thermocyclers. Please use only half and full-skirted 96-well plates. If you intend to submit multiple
96-well plates, please inform us in advance so we can check if blocks on thermocyclers are available. It is
assumed that samples are filled on the plates column-wise (A1-H1, A2-H2…A12-H12) for 96-well plates.
If you intend to use 384-well plates, please check with us for further instructions.
