Dissolve ingredients (except NaHCO3), bring to a boil, cool down to room temperature while gasing
the medium with 80% N2 + 20% CO2, and add the bicarbonate. Equilibrate the medium with the gas to
reach a pH of 6.8. Dispense the medium in anaerobe tubes under N2 + CO2. After autoclaving add
fumarate from an anaerobic filter sterilized stock solution (16% w/v Na2-fumarate; 0.5 ml/10 ml
medium). Check medium pH which should be around 6.8.

Prepare the medium anaerobically under 80% H2 + 20% CO2 atmosphere. For incubation use the same gas
mixture at two atmospheres of pressure. If the medium is being used without gas mixture overpressure
then adjust pH with a little hydrochloric acid. Final pH should be 7.0 for strains DSM 1497, DSM
1537, DSM 2067, DSM 2279, and DSM 2373; 6.8 for strain DSM 2095, and 6.5 for strain DSM 1498. For
DSM 2373 increase the amount of trypticase to 6 g/l.
For DSM 14042 prepare the medium under 80% N2 + 20% CO2 gas atmosphere. After sterilization add
0.05% methanol from a sterile, anoxic stock solution prepared under N2. For incubation use the same
gas mixture at two atmospheres of pressure. The final pH should be 7.0.

Medium 385: DESULFOBACTERIUM CATECHOLICUM MEDIUM
Use medium 193 but lower the amount of NaCl to 1 g/l and of MgCl2 x 6 H2O to 0.4 g/l. Sodium acetate
is replaced by filter-sterilized sodium benzoate (0.37 g/l) plus catechol (0.055 g/l). Use only 2.5
g/l of sodium bicarbonate!
Add 1 ml/l selenite- and tungstate solution (0.5 g NaOH, 3 mg Na2SeO3 x 5 H2O, 4 mg Na2WO4 x 2 H2O,
1 l distilled water). Final pH of the medium should be 6.9 - 7.1.
