p r e s e r v e d o p a m i n e f u n c t i o n i n th e n i g r o - s t r i a t a l
pathway of the same animals used in the behavioral
study.
I n t h e b e h a v i o r a l s t u d y , f i v e g r o u ps o f a d u l t m a l e
C 5 7 B 1 / B 6 m i ce r e ce i v e d i m p l a n ts o f o s m o t i c
minipumps containing a 14-day supply of either vehicle
o r o n e o f t h r e e c om p o u n d s s u p p l i e d b y C o m p a n y
(designated 1, 2, and 3). Three days later the animals
r e c e i v e d t w o b e h av i o r a l a s s a y s o f s tr i a t a l f u n c t i o n
(Open Field test, Tail Hanging). Immediately following
t h e s e t e s ts t h e a n i m a l s r e ce i v e d s y s te m i c i n j e c t i o n s o f

m o u s e m o d e l o f Pa r k i n s on ’ s d i s e as e , a s w e l l a s t o p r e v e n t
t h e m o t o r d e f i ci t s t h a t o c c u r i n t h i s m o d e l .
Methods and Experimental Design
Design
F i f t y a d u l t m a l e C5 7 B l 6 mi c e we r e ra n d o m l y d i v i de d
i n t o 5 g r o u p s . Al l w e r e t e s t e d i n t h e O p e n - F i e l d a n d T a i l H a n g i n g t e s t s o f s t r i a t a l f u n c t i o n . T h e O pe n F i e l d t e s t i s
t h e m o s t f re q u e n t l y u s e d b e h a v i o r a l a s s a y i n t h e M P T P
mouse model, and is thought to be sensitive to loss of
dopaminergic input to the striatum from the substantia
nigra. The Tail Hang test is known to be sensitive to direct
striatal damage.

Subjects
T h e m i ce w e re C 5 7 B 1 /B 6 t y p e a n d w e r e o b t a i n e d f r om
J a c k s o n L a b o r a t o r i e s . T h e y w e re 8 -1 2 w e e k s o l d u p o n
a r r i v a l a t t h e N e u ro D e t e c t i v e a n i m a l c o l o n y . T h e y w e re
allowed to acclimatize for 10-14 days prior to any
e x pe r i me n t a l m a n i p u l a t i o n s . A l l m i ce w e r e h ou s e d i n gr o u p s
o f 3 - 4 p e r c a g e a n d w e r e p r o v i de d wi t h a d - l i b i t u m a c c e s s t o
f o o d a n d w a t e r . T h e y we r e m a i n t a i n e d o n a 1 2 h r : 1 2 h r
l i g h t / d a r k c y c l e . T h e a n i m a l s w e re ra n d o m l y a s s i g n e d t o

Procedures
A f t e r t h e m i c e h a d b e e n a c c l i m a t e d t h e y we r e
r a n d o m l y a s s i g n e d t o o n e o f t h e 5 g ro u p s ( s e e a b o v e ) .
E a c h m o u s e w a s t h e n g i v e n a n i n i t i a l O p e n - Fi e l d t e s t a n d a
T a i l H a n g i n g t e s t ( d e s c r i be d b e l o w ) . F o l l o w i n g t h e s e te s t s
e a c h m o u s e i n 4 of t h e g r o u p s w as i m p l a n t e d w i t h a mi n i o s m o t i c p u m p . T h e i m p l a n t s we r e pe r f o r me d u n de r
i s o f l u r a n e a n e s t h e t i c . T h e m i n i - p u mp s c o n t a i n e d o n e o f
three compounds: 2 (3 mg/kg/day in DMSO vehicle), 1 (3
m g / k g / d a y i n D MS O v e h i c l e ) , o r 3 ( 1 m g / k g / d a y ) ; o r v e h i cl e
o n l y . T h e i n f u s i o n s w e re g i v e n a t t h e r a t e o f 0 .2 5 u l / h r f o r
14 days. Three days following implantation of the pumps,
a n d f o l l o w i n g b e h a v i o r a l t e s t i n g (s e e D e s i g n ) , t h e m i ce
r e c e i v e d a s u b c u t a n e o u s i n je c t i o n o f 4 0 m g / k g M P T P . T e n t w e l v e d a y s f o l l o wi n g M P T P i n je c t i o n s e a c h m o u s e w as
tested again in the Open-Field and Tail Hanging tests (see
b e l o w ) . T h e s e t e s t s w a s r e p e a t e d a t 3 0 - 4 0 d a y s p os t MPTP, and again at 50-55 days post MPTP injections.

NEURON COUNTING:
B e t w e e n 5 5 a n d 6 0 d a y s p o s t M P T P i n j e c t i on s , t h e
m i c e w e r e s a c r i f i ce d b y a n o v e r d os e o f S o d i u m N e m b u t a l .
T h e i r b r a i n s w e re p e r f u s e d w i t h ph os p h a t e b u f f e re d s al i n e
f o l l o w e d b y L a n a ’ s f i x a t i v e ( p a r a f o r m a l d e h y de a n d p i cr i c
a c i d ) . T h e b r a i n s w e r e re m o v e d a n d p l a c e d i n L a n a ’s
fixative for 7 – 10 days. The brains were then cut
c o r o n a l l y a t 5 0 µ m u s i n g a v i b r a t o m e , a n d t h e s e c t i on s
stained with an antibody to tyrosine hydroxylase (TH), the

were selected for cell counting in the SN and the VTA.
These sections are at the midpoint of the rostral half and
the midpoint of the caudal half of SN respectively. Each THl a b e l e d ce l l t h a t w a s c l e a r l y v i s i b l e a n d h a d b e t w e e n 2 a n d
6 n e u r i t e s w a s c on s i de r e d a n e u r o n a n d c o u n t e d . A n

average was obtained for the four sections. Separate

Results
4.1. Open Field test
Prior to MPTP lesioning there was no significant
d i f f e r e n ce a m o n g t h e 5 g r o u ps i n t h e o p e n - f i e l d a c t i v i t y
t e s t i n d i s t a n c e t ra v e l e d o r n u m b e r o f r e a r s , a l t h o u gh t h e 3
group tended to be the least active and Company compound
g r o u p s t h e m os t a c t i v e i n o v e r a l l l o co m o t o r a c t i v i t y ( p . A 1 ) .
T h e s e t e n de n c i e s c o u l d r e f l e c t s l i g h t d i f f e re n ce s i n o v e r a l l
r e s p o n s i v e n e s s t o t h e d i f f e re n t d r u gs a f t e r 3 d a y s , a b s e n t
any lesion effect.
