Promega began producing restriction enzymes in 1978. Each of
our enzymes represents the cumination of many years experience
manufacturing high-quality products for molecular biology research.
Have more confidence cloning with Blue/White Cloning
Qualified Enzymes
We test enzymes that are commonly found in vector multiple
cloning sites by the Blue/White Cloning Qualified Assay, originated
at Promega. This functional assay provides a high level of quality
control, especially for blunt-end cutters. Without this level of
integrity, false positive or irretrievable inserts may result from
contaminating nuclease activities or other impurities in the enzyme.
This sensitive assay allows detection of a single base deletion
within a restriction site. The Promega Blue/White Cloning Assay
uses a 5-fold excess digestion of a single-site vector, which is then
re-ligated and transformed into JM109 cells. After growth on X-Gal/
IPTG/Amp plates, the percentage of white colonies—representing
false positives—is calculated.

Genome Qualified Enzymes are tested on chromosomal DNA
templates embedded in agarose plugs. All genome-qualified
enzymes yield clear and complete digestion patterns in pulsed-field
gels stained with ethidium bromide.
