This functional assay provides a high level of quality control, especially for blunt-end cutters. Without this level of integrity, false positive or irretrievable inserts may result from contaminating nuclease activities or other impurities in the enzyme. This sensitive assay allows detection of a single base deletion within a restriction site. After growth on X-Gal/ IPTG/Amp plates, the percentage of white colonies—representing false positives—is calculated.
