Description
Indicating Whatman FTA cards are designed for the collection of clear
biological samples including buccal cells for DNA isolation. Cells are
captured on the foam tipped applicator by swabbing the oral mucosa
and subsequently transferred to the Indicating FTA Card. Upon contact,
the proprietary chemical formulation lyses cell, denatures proteins and
binds DNA to the card matrix. The captured DNA is then protected from
degradation during long term room temperature storage. The pink
indicating dye will turn white to mark the location of the sample. To
prepare the indicating FTA card for use with downstream applications,
allow the card to air dry at room temperature after applying the
biological sample.
Precautions
Handling: Follow universal precautions for handling of biological
samples. Always wear gloves when handling indicating FTA cards and
avoid touching the pink sample collection area of the card. DNA yields
may be improved by having the person donating the sample avoid
eating, drinking, or smoking 30 minutes prior to sample collection. Avoid
rubbing the foam applicator on the FTA card as it will damage the
FTA matrix.
Processing Samples with the kit
1. Open the kit (Figure 1). Lay the contents of the package out on a
clean and dry surface. The kit contents are as follows;

Peelable barcodes, × 2

Sterile Foam Tipped Applicator, × 1

Return mailing envelop – 8.7 inches × 4.3 inches
(22 cm × 11 cm)
e.
Multi-barrier pouch, 4.4 inches × 6.5 inches (11 cm × 16.5 cm)
f.
Nitrile gloves – 1 pair, size Large
g.
Tamper evident tape
h.Desiccant
i.
The above components are assembled inside a nylon
pouch, 9 inches × 12 inches (22.8 cm × 30.5 cm)
Note: Download, print and complete the demographics form. The
demographic form is available for download at
www.gelifesciences.com/demographics. Apply a peelable
bar code to the form for traceability back to the FTA card.

2. Place the Indicating FTA Card on a clean, dry, flat surface. Label the
card with a unique identifying name, number, etc.
3. Remove one Sterile Foam Tipped Applicator (Part No. WB100032)
from the protective packaging according to instructions on the
packaging.
4. Holding the plastic handle
of the applicator, place the
foam applicator in the mouth.
In order to soak up as much
saliva as possible, run the
foam head along the gum line
and under the tongue. After
you have made the foam
applicator wet with saliva,
rub one side of the foam
applicator on the inside of the
cheek for 15 seconds (Figure
2). Repeat using the opposite
side of the foam applicator for
the other cheek. Remove the
applicator from the mouth.
5. Carefully lift the paper cover of the
Indicating FTA Card to expose the
pink sample collection area. Press
the flat, circular foam applicator
within the circled area. Without lifting
the foam from the card and while
applying pressure, rock the foam
applicator from side to side three
times (Figure 3 and Figure 4).
Turn the applicator over and repeat
with the other side within the same
circle. The sample collection area
will turn white indicating the transfer
of sample. After the foam tip has
touched the card, do not re-insert it
into the mouth.

6. Discard the applicator according to local laboratory procedures.

7. Position the Indicating FTA Card
for drying by supporting the
sample collection area with the
paper cover as shown
(Figure 5). Allow the card
to completely dry at room
temperature.

8. If buccal cells are to be applied to more than one sample collection
area, use a new applicator (Part No. WB100032) and repeat steps
1–6 on a different sample collection area or on a new card.
9. After the Indicating FTA Card is dry, the sample can be processed
immediately or packaged for transport or storage.
10. To transport or store the Indicating FTA Card, place the Card with a
Desiccant Packet (Part No. WB100003) into the Multi-Barrier Pouch.
Apply a peelable bar code to the outside of the Multi-Barrier
Pouch (Part No. WB100036 or WB100037). Remove the liner from
the adhesive strip on the Multi-Barrier Pouch and seal by folding
the flap. Place the tamper evident tape across the seal of the
mailing envelope. Insert the Multi-Barrier Pouch and the completed
demographics form into the Return Mailing Envelop. Seal the
mailing envelop as necessary.
The stored Indicating FTA Card is now ready for shipment or
storage. Retain the envelope at room temperature.
Note: The components of the kit are for single use.
Storage of samples on indicating FTA Cards
Biological samples applied to Indicating FTA Cards should be archived
at room temperature in a Multi-Barrier Pouch with a desiccant packet.
