The mixture contains three standard oligonucleotides. The compounds are listed in the following table together with their molecular weights. The standard oligonucleotides are supplied in five tubes per package. Each tube containing 25 pmoles Oligo 12, 125 pmoles Oligo 20 and 500 pmoles Oligo 30. The quantity of substance allows 5x100 calibration points. We recommend storing the oligonucleotides on arrival at less than 0 °C. Dissolved samples should be aliquoted and frozen. We do not recommend refreezing samples after thawing. Risk and safety information This kit is not subject to classification according to the calculation methods of the “General Classification Guideline for Preparations of the EU” as issued in the latest valid version. The product is not subject to identification regulations under EU Directives and the Ordinance on Hazardous Materials (GefStoffV). When used and handled according to specifications, the product does not have any harmful effects according to our experience. All materials may present unknown hazards and should be used with caution. Observe the general safety regulations when handling chemicals. Sample preparation procedure (recommendation) Preliminary remarks Poor sample preparation will degrade sensitivity, yield low resolution and poor reproducibility. The generation of ions through MALDI depends on the production of a suitable composite material, consisting of the matrix substance and the analyte. For best results use only chemicals of highest available purity. Chemicals and materials required 3-HPA ACN AHC 3-Hydroxypicolinic acid (1 g: #201224; 5 g: #203070) Acetonitrile Diammonium hydrogen citrate (100 mg/ml) in ultra pure water Equipment and tools required: centrifuge, shaker, ultra sonic device, pipettes, pipette tips 1. Preparation of standard solution Dissolve a sample of oligonucleotide calibration standard (contents of a tube) in 100 µl ultrapure water and shake / whirl mix for several seconds (COligo 1 = 0.25 pmol/µl, COligo 2 = 1.25 pmol/µl, COligo 3 = 5 pmol/µl). 2. Preparation of matrix solution a) for standard steel targets: - produce a solution of 3-HPA in ultrapure water / ACN (1:1 v/v) with a concentration of 50 mg/ml (if necessary, assist solution process by help of ultrasonic device) - mix solution of 3-HPA with AHC (10:1 v/v) b) for AnchorChipTM targets: - mix 10 mg 3-HPA and 10 µl of AHC solution in 990 µl ultrapure water (if necessary, assist solution process by help of ultrasonic device) 3. Preparation onto MALDI TOF-Target - Apply 1 µl matrix solution onto the target and allow drying at room temperature. Add 1 µl standard solution and allow drying at room temperature once more.
