AHN PCR-Micro Test Tubes basically withstand the thermal and mechanical strain of
being heated in a thermoblock or being subjected to high-speed centrifugation. It is

essential to select the correct Micro Test Tube like AHN PCR-Tubes. They are availabe in
2 volumes, 0.2 ml and 0.5 ml, with optimized clear transparency. These tubes are
available in different colours and are manufactured in clean rooms under DNA-free,
RNase-free, ATP-free and pyrogene-free conditions. Optimized for all thermocyclers AHN
customer can choose between 2 designs with flat caps or domed caps. Autoclavable when

The efficiency and flexibility of AHN
Biotechnologie PCR tube-strips is
ensured by the fact that they all are
manufactured from Polypropylene.
The recovery of sample products is
increased
in
comparison
to
experiments
carried
out
using
polycarbonate equipment, and the risk
of biological contamination is reduced.
Safety and flexibility are also
increased as polypropylene equipment
can be autoclaved and sealed using
many different methods.

5.4 qPCR-Capillaries
Safer, easier, cheaper qPCR
Free of human DNA, certified free of DNase,
RNase and PCR inhibitors
No precipitates
Plastic capillaries from AHN Biotechnologie
do not require the use of BSA in the reaction
mix. In this way precipitates, that can disturb
the optical detection, are avoided.
More optimal reaction
The reaction characteristics are more
optimal as PCR components like TAQ
polymerase, MGCl2, primers, probes and
target DNA do not adhere to the optical
plastic in the way they adhere to glass. The
result is more active components in solution
and therefore a more efficient PCR, using
less target DNA.
Non breaking plastic capillaries 50 l
AHN Biotechnologie has developed a proprietary
solution to frequently breaking Real-Time PCR glass
capillaries. When using these optical plastic capillaries
you avoid the risk of breaking, contamination and injury.
Higher reaction volume
Because of the special design of the tubes,
they can hold up to 50 l reaction mix, which
allows you to add more sample DNA, and
therefore allows you to increase the
sensitivity. This is especially very useful
when detecting low copy numbers of DNA.
Post reaction sample recovery
The special design of the innovative AHN
qPCR-capillaries allows you also to recover
sample after Real-Time PCR with a 10 l
pipette.

5.5 qPCR-Capillaries

AHN qPCR-Capillaries ® User Guide
Sample preparation / PCR mixture preparation
Samples for PCR can be handled and prepared according to the existing protocol at customer site.
No changes have to be made using AHN polycarbonate capillaries instead of the original glass
capillaries. No changes have to be made preparing the PCR Mix.
It is possible to use AHN polycarbonate capillaries for PCR volumes from 10 µl to 50 µl without
significant changes in the PCR modes (for details see point PCR mode).
Handling capillaries and caps
For better/easier handling of our capillaries and caps we provide a “Transfer-Pin” for taking the
capillaries and the caps out of the rack-box. The Transfer-Pin is part of each sales unit. If you are
used to work without a “Transfer-Pin”, you should use protective gloves to protect you PCRreaction from contamination like DNAse.
Avoid wetting the rim when pipetting the sample into the capillaries. If the rim of the
capillaries is wetted the closure of the caps could be a problem. Therefore pipet direct and
straight in the capillary. For sealing the capillaries with the caps, please use the transfer pin, to
transfer caps from the rack-box to the polycarbonate capillary. Ensure that each capillary is closed
tightly by checking it visually.
Performing PCR / PCR mode (protocol)
Our AHN polycarbonate capillaries have a bigger diameter, compared to the 20 µl glass capillaries.
To guarantee effective heat transfer from the outside of the capillaries to the middle of the PCR
reaction, despite the bigger diameter, it might be necessary to increase the “hold-times” at
denaturing and annealing. For this reason we recommend to program your Cycler mode with
longer hold times for the denaturing and annealing steps. We suggest to program at least 7
seconds at 95°C for the denaturing step and 5 seconds for the annealing step. To short denaturing
and annealing times will lead to a complete failure of your PCR, or at least to much higher CTtimes.
If you are already using a mode with both times higher than 7 seconds, no changes have to be
made normally. Sometimes it might be better to increase the existing times by 2 seconds each.
As the effectiveness of denaturing is also depending on the GC-content of your DNA, it might be
even better to prolong denaturing up to 10 seconds (note: only in case of no PCR signal with a
denaturing time of 5 seconds).
Sample recovery after PCR
Some customer wants to analyse the PCR reaction on a gel. For that purpose the cap can be
taken from the capillary. The recovery of the sample is principally possible using pipettes with a 10
µl tip. In some cases the pipet tips are too broad. In this case you can open the capillaries and spin
the solution into a clean reaction tube.
