Brief Description of the Institution CIC bioGUNE (Center for Cooperative Research in Biosciences) is a non-‐profit biomedical research organization located at the Science and Technology Park of Bizkaia. The Center has invested over 40 million € in state-‐of-‐the-‐art research infrastructures, including molecular imaging, genomics, proteomics, metabolomics, NMR, electron microscopy, optical microscopy, X-‐ray diffraction, computer and animal facilities, to support the research activities of their staff and students. Every year, the Centre commits more than 10 million € to research and dedicates 450 000 € to PhD training. It employs 22 faculty investigators, more than 85 postdocs, technicians and engineers, and provides training opportunities to 23 PhD students each year. The Structural Biology Section is divided into three platforms. The NMR spectroscopy platform is equipped with a 600MHz and a 800MHz Bruker Avance III spectrometers. The electron microscopy platform uses a JEM-‐2200FS/CR and a JEM-‐1230 transmission electron microscopes. The X-‐ray crystallography platform is equipped with a X8 Proteum Integrated System, a liquid-‐handling robot (Tecan EVO-‐100), a nanodispenser robot (Mosquito), a Crystal Farm 400 Imaging System (Bruker) and vibration-‐free rooms (at 4 ºC and 18 ºC) for crystallisation. The Centre has a dedicated IT Structural Biology support unit catering to our intensive data processing and storage requirements. This unit holds a high performance cluster system (with more than 400 cores) for high throughput analysis. 3. Research / Project Description Living cells constantly recycle receptors, proteins and lipids with a direct impact on nutrient uptake, re-‐sensitisation to environmental signals, immune surveillance and waste management. Failure to recycle results in reduced signalling, oxidative stress, protein mislocalisation and aggregation, which are pathological hallmarks of many cardiovascular, cancer, and neurodegenerative diseases. Endosomes are key recycling compartments where the biosynthetic and endocytic pathways intersect. Here, the fate of sorting receptors is directly linked to their selective recruitment into tubulo-‐vesicular carriers. Our understanding of receptor recycling in a motif-‐dependent manner, the formation of such tubulo-‐vesicles and the functional components defining their architecture remains very limited. Retromer is a novel protein coat complex with a central role in multiple receptor-‐sorting events in the endosomes. Retromer combines receptor recruitment with membrane tubulation properties, but unlike other classical coats, it does so through a single-‐layer or “cage-‐free” organization. This laboratory is interested in understanding how retromer components form a coat around tubulo-‐vesicles, how transmembrane receptors are selectively recruited to control essential cell processes, and how certain pathogens subvert and co-‐opt these interactions. 5.
