The products described in this document are intended
to be used for the clarification of mammalian and
microbial cell cultures. Sartorius shall not assume any
liability if Sartolab® products have been subjected
to improper use. The use of this product for any other
applications or in another manner than described
in this manual may result in personal injury or
malfunction of the product. Please follow the
instructions in this user manual to ensure your own
safety and to obtain optimal filtration results.

Directions for Use
The purpose of this document is to outline the
­appropriate method for using the Sartolab DY 0.15L
kit for body feed filtration.
The device described in the document is intended to
be used for the clarification of animal and microbial
cell cultures and other fluids of biological origin.
Introduction
The addition of filter aids like diatomaceous earth
(DE) to increase the total throughput in clarification
applications is used in many different industries.
DE is a highly porous, insoluble material and can
be used as a spacer between solid particles. While
these p
­ articles are entrapped in a filter cake, liquid,
including dissolved substances, can easily flow
through the filter cake by passing through the porous
structure of the DE particles.
The Sartolab DY 0.15L kit is appropriate for the
­clarification of cell cultures containing a maximum
of 12% wet biomass.

Connect your Sartolab DY 0.15L filter unit to
a vacuum system, e.g. Microsart® e-jet pump,
order no. 166MP-4 (vacuum max. 0.4-bar).

Take a 150 mL sample of your cell culture.

Optional step: When using mammalian cell
lines producing therapeutic antibody product,
it is recommended to adjust your sample to a
pH of 5 using acetic acid (2M or 30%) and to
stir the suspension for a minimum of 45 min.
before filtration. This will allow correct precipitation of small particle portions. Gentle mixing
with a magnetic stirring bar is recommended.

Add your sample to 1 bottle of DE and vortex
to homogenize. You need to perform this
procedure in a ventilated area, such as under a
fume hood, to avoid inhaling dust particles.

Add your homogenized suspension to the
Sartolab DY 0.15L filter unit and turn on the
vacuum pump. Wait until your filter unit is
completely drained.

Optional step: To avoid any further precipitation reaction after filtration, adjust the filtrate
pH back to pH 7 using NaOH (1M).

Discard the top half of the Sartolab filter unit
containing the cell and diatomaceous earth
mixture in accordance with your local legal
regulations for waste disposal.

Return of Used Products
All used materials must be properly sterilized prior to
shipping to Sartorius. This will enable our staff to
handle them with minimal risk during inspection of

representative) be completed prior to shipping of
used filter materials.
