Briefly discuss the major difference between the classical Raman and Surface enhanced Raman What is the minimal Raman and Surface‐enhanced Raman. What is the minimal number of molecules that SERS can detect? In general, surface‐enhanced Raman analysis is more sensitive than classical l f h d l h l l Raman analysis. Surface‐enhanced Raman spectroscopy allows for more detailed analysis of the material being studied, because it can study the surface of the material rather than the bulk of the material. Surface‐ enhanced Raman spectroscopy can detect a single molecule. 2. If you are hired as a conservator by a museum and is free to choose your first project using SERS or related equipment what will be the your first project using SERS or related equipment, what will be the first archeological or art object that you wish to look into? Why? The first art object I would choose to study using advanced imaging h f b ld h d d d techniques would be a painting called The Garden of Earthly Delights, painted by artist Hieronymus Bosch between 1490 and 1510. The painting is in fairly good condition, but has still been damaged by the passage of time. Being able to used advanced imaging techniques on the painting could allow for p paint and drawing analysis, and the original colors and lines of the painting g y , g p g could be discovered. Using that information, the painting could be restored to its original glory. Bosch displays extreme skill in small detail, and being able to see what the painting once looked like would be incredible able to see what the painting once looked like would be incredible. 3. You have seen the restored painting “For to be a farmer’s boy”, which completely changed the interpretation of that work. Leonardo da Vinci’s painting The Last Supper d d ’ h underwent intense restoration d between 1978 and 1999 in order to help restore the color and detail of the original fresco, which had been badly damaged by previous, unsuccessful restorations. While the restoration does not change the interpretation of the work as in For to be a Farmer’s Boy, it is important to ensure that the work will be around for future generations to enjoy. Techniques such as infrared g j y q reflectoscopy and microscopic core sampling were used to discover more of the original details and colors. Pictures are on the next two slides. This is how the painting looked before its restoration in 1978. hi i h h i i l k db f i i i 9 8 <http://en.wikipedia.org/wiki/File:Leonardo_da_Vinci_‐_Ultima_cena_‐_ca_1975.jpg> The painting as it appears today, after restoration. <http://en.wikipedia.org/wiki/File:%C3%9Altima_Cena_‐_Da_Vinci_5.jpg> Homework 2 100929 Viray 2 1. Briefly discuss the major difference between y j the classical Raman and Surface‐enhanced Raman What is the minimal number of Raman. What is the minimal number of molecules that SERS can detect? • The major difference between classical Raman and S f Surface‐enhanced Raman is that SERS h dR i th t SERS is normally i ll done when object has high amounts of gold and silver as it results “in an increase in the electric fields il it lt “i i i th l t i fi ld surrounding the metal,” (Wikipedia). Using these i t iti SERS can measure a signal up to a 10^11 intensities, SERS i l t 10^11 more than Classical Raman. (single molecule) 2. If you are hired as a conservator by a museum and is free to choose your first project using SERS or related equipment, y p j g q p , what will be the first archeological or art object that you wish to look into? Why? • The first archeological object I would look into using SERS or other related equipment would have to be cave paintings, other related equipment would have to be cave paintings, whether they be from Neanderthals or even the earliest Homo Sapiens. Cave paintings have always fascinated me, I would like to look at further symbols our ancestors had ld lik t l k t f th b l t h d painted before us. Even more so, using this technology we could figure out what type of natural pigments they used to g yp pg y create their “make‐shift” paint. Perhaps they used different plants or flowers, or maybe even more concept things such as organisms Nonetheless we would be able to see what tools organisms. Nonetheless, we would be able to see what tools they used to create these dyes, something that must have taken skill and effort on their part. 3.
