PRODUCT DESCRIPTION:

The C 18 LithTips Sample Prep Kits contain tips and reagents for
reversed-phase solid phase extraction for protein sample clean-up
and for spotting of peptides and small proteins from samples for
MALDI mass spectrometry. These kits are effective in concentrating
and desalting peptides and small proteins, resulting in mass
spectra with better S/N and reduced salt adducts. The Sample Prep
Kits contain C 18 LithTips for 1-10 µL sample volumes, 10 mL
Sample Reconstitution and Rinse solution, 10 mL Equilibration
Solution, and 10 mL Elution Solution. The C
18 monoliths are
immobilized on the inner surface of a pipette tip, providing a large
surface area for sample capture. In addition, the central portion of
the monolith lining the tip is hollow, eliminating backpressure
problems during pipetting.
PROTOCOL:
1) Add 10-20 µL of the Sample Reconstitution and Rinse Solution to
the solid sample and sonicate briefly to dissolve the sample.
2) Wash and equilibrate a C
18 LithTip using the Equilibration Buffer.
Add 25 µL of buffer to a clean vial and then perform 10 cycles of
aspiration and expulsion of the buffer using the micropipette set at
10 µL.
3) Rinse the C 18 LithTip using Reconstitution and Rinse Solution.
Add 25 µL of buffer to a clean vial and then perform 10 cycles of
aspiration and expulsion of the buffer using the micropipette set at
10 µL.
4) Load the sample into the C
18 LithTip by aspirating and expelling
the sample 5-10 times within the sample vial.
5) Wash the sample on the C
18 LithTip with GPR Rinse Buffer for
MALDI.
a. Add 25 µL of buffer to a clean vial and then perform 10 cycles of
aspiration of the buffer using the micropipette set at 10 µL.
6) Elute the sample with 1.5 µL of Elution Solution, by spotting the
1.5 µL directly onto the MALDI target, pre-spotted with 1.0 µL
MALDI matrix (e.g. CHCA) and allowing to air-dry.
7) Load the MALDI target into the mass spectrometer and begin
mass spec analysis.
PRODUCT CARE AND STORAGE:
Product is shipped at ambient temperature and stored at room
temperature. Recommended for use within 2 years.
