Phosphatidylserine Receptor (clone 217)

T
he culmination of apoptosis in vivo is phagocytosis of cellular corpses. During apoptosis, plasma membrane phospholipid asymmetry is lost, exposing phosphatidylserine (PS) externally.
Phagocytosis of apoptotic cells can be inhibited stereospecifically by PS and it’s structural
analogs, but not by other anionic phospholipids, suggesting that PS is specifically recognized.
A novel gene, cloned by phage display, appears to recognize PS on apoptotic cells. This gene, when
transfected into B and T lymphocytes, enables them to recognize and engulf apoptotic cells in a PSspecific manner. Flow cytometric analysis using antibodies to the PS receptor (clone designation MAb
217) suggests that the protein is expressed on the surface of macrophages, fibroblasts and epithelial
cells. The antibody, like PS liposomes, inhibited their phagocytosis of apoptotic cells and, in
macrophages, induced an anti-inflammatory state. The candidate PS receptor is highly homologous
to genes of unknown function in C. elegans and D. melanogaster, suggesting that the PS recognition
on apoptotic cells during their removal by phagocytes is highly conserved throughout phylogeny.
