Introduction 3.2 Materials The RK 16/26 (Code No. 18-8793-01) and RK 50 (Code No. 18-8790-01) packing reservoirs are designed for use with the XK columns. Each unit consists of a cylinder with a conical adaptor, which screws into the respective XK column, and a screw cap, see exploded diagram below. The pressure limit is 0.5 MPa (5 bar). The reservoir can also be used as an eluent container. The materials from which the individual parts of the packing reservoir are made are listed in the spare parts list below. All these materials may come in contact with the gel suspension or eluent during the packing procedure. 2. Unpacking The packing reservoir is delivered complete with flanged capillary tubing with M6 connectors. Please check the delivery against the packing list. 3. Description 3.1 Functional design Please refer to the exploded diagram below for details. Cylinder (4). The cylinder is a transparent plastic tube threaded at each end and graduated at 20 ml intervals to 300 ml or 800 ml. Cylinder dimensions are as follows: 3.3 Chemical resistance The packing reservoirs may be used with aqueous solutions and nearly all organic solvents commonly used in liquid chromatography of macromolecules, with the following few exceptions: chlorinated hydrocarbons, acetone and other ketones, aliphatic esters, phenol, ammonia, and concentrations of alcohol greater than 20%. Solutions containing more than 10% NaOH, 10% HCl or other strong mineral acids, 5% acetic acid and trichloro-acetic acid, should not be used. The reservoir can be used at temperatures up to 50 °C and at pressures up to 0.5 MPa (5 bar, 70 psi). It may be sterilized with ethylene oxide. It should not be autoclaved. 4. Installation 1. The adaptor screws onto the cylinder and into the respective XK column. There are 3 conical adaptors, one for each XK column series: XK16, XK26 and XK50. The sealing against the columns is obtained via the sealing ring (1). The volume of the conical adaptors is 50, 70 and 100 ml for the XK16, XK26 and XK50 columns, respectively. Cap (5). The screw cap screws onto either end of the cylinders RK16/26 or RK50. The cap carries a M6 nipple with capillary tubing (6) for connection to valves, connectors and pump. 2. Select the appropriate conical adaptor (2) and screw it onto the XK column. Check that the seal ring (1) and the O-rings (3) are in place. 3. Screw the appropriate cylinder (4) onto the conical adaptor. 4. Packing with the reservoir is described in the package TM insert included in every package of Sephacryl HR (High Resolution) gels and in “Gel Filtration, Principles and Methods”, a handbook available from GE Healthcare. 5. Cleaning Suitable cleaning agents are soapy water or laboratory detergents. 6. Spare parts The exploded diagram shows the correct locations of the different parts of the RK packing reservoirs. For replacements, please order according to the spare part list below using the appropriate code numbers. Please note that the code numbers for both packing reservoirs RK16/26 and RK50 are included in the same table.
