TOTAL ALKALINE PHOSPHATASE AND ISOENZYME LEVELS IN PERSONS WITH Hb - GENOTYPE AA AND SS RESPECTIVELY CHIZEA, DANIEL CHfNEDU /1/ljTR/[ No: 96/0241 A RESEARCH PROJECT SUBMITT~D TO THE DEPARTMENT OF CHEMICAL SCIENCES, COLLEGE OF NATURAL SCIENCE, UNIVERISTY OF AGRICULTURE, ABEOKUT A, OGUN STATE IN PARTIAL FULFILMENT OF THE REQUIREMENT FOR THE AWARD OF BACHELOR OF SCIENCE (B.Se) (CHEMISTRY / BIOCHEMISTRY) OF THE UNIVERSITY OF AGRICULTURE, ABEOKUT A, NIGERIA. Abstract The serum ALP and isoenzyme levels are studied with a view to, at least, being able to fashion out a viable predictive means of diagnosing crisis condition in a sickler. Alkaline phosphatases hydrolyse a number of ester orthophosphates at alkaline pH, but the substrates acted upon physiologically, are unknown. Sera of persons with Hb-genotype AA and SS respectively, were obtained by centrifuging fresh whole blood. The appropriate concentrations (p-nitrophenylphosphate) of enzyme and substrate were then mixed, yielding p-nitrophenol with yellow color. This is. measured at 405nm with a spectrophotometre. Findings revealed that there is a statistical difference in the level of enzyme activity between the two genotypes.
