Abstract
The serum ALP and isoenzyme levels are studied with a view to, at least,
being able to fashion out a viable predictive means of diagnosing crisis
condition in a sickler. Alkaline phosphatases hydrolyse a number of ester
orthophosphates
at
alkaline
pH,
but
the
substrates
acted
upon
physiologically, are unknown. Sera of persons with Hb-genotype AA and
SS respectively, were obtained by centrifuging fresh whole blood. The
appropriate
concentrations
(p-nitrophenylphosphate)
of
enzyme
and
substrate
were then mixed, yielding p-nitrophenol
with
yellow color. This is. measured at 405nm with a spectrophotometre.
Findings revealed that there is a statistical difference in the level of
enzyme activity between the two genotypes.
