Functional Validation
Anti-human PD-1 inhibits the binding of PD-L1
The graph shows two sets of data, PD-1 binding to PD-L1 (cyan lines) and PD-1/PD-L1
interaction blockade by anti-PD-L1 (purple lines). PD-L1-Fc was coated at 1 µg/mL and
increasing concentrations of PD-1-Fc (cyan diamonds, top scale) or paired PBS wells
(cyan circles, top scale) were incubated overnight. The bound PD-1-Fc was detected
using biotin anti-mouse PD-1 and Avidin-HRP followed by TMB. After verifying PD-1/
PD-L1 binding, coated PD-L1-Fc was pre-incubated with increasing concentrations
of anti-mouse PD-L1 (clone 10F.9G2, purple squares, bottom scale) or isotype control
(clone RTK4530, purple circles, bottom scale), followed by detection as described. Clone
10F.9G2 inhibits PD-1/PD-L1 interaction in a dose-dependent manner.
Treg induction by anti-mouse DR3 (TNFRSF25)
C57BL/6 mice were IP injected with 100 μg LEAF™ anti-DR3 (clone 4C12) (3 mice) or
LEAF™ Armenian Hamster IgG (3 mice). On day 0, 3, 6, 7, and 11, the blood was drawn
and stained with CD4-APC followed by intracellular staining with FOXP3-PE. The data
was analyzed as a percentage of FOXP3 cells of total CD4+ cells. Error bars are ± SD.
