Sartobind® 75 mL, 4 mm and 150 mL, 8 mm bed height Void-volume-optimized membrane chromatography capsules for pilot scale Benefits – Applicable for flow-through polishing including bind & elute applications – Increased dynamic binding capacity at 10% breakthrough – Small elution volumes compared to non-optimized capsules – Plug and play pilot scale device – Disposable or reusable Product Information Sartobind 75 and 150 mL capsules are intermediate sizes of void-volume optimized capsules with 4 and 8 mm membrane bed height. For the capture of large proteins or viruses it is important to achieve a steep breakthrough curve and have the smallest possible elution volume. The optimized construction of the capsule makes it ideal for both removing contaminants | impurities via flow-through polishing and for bind and elute applications. The SingleSep® product line has been a standard for flow-through applications, but these capsules are not void-volume-optimized. The new 75 mL version is optimized and will replace the 5” SingleSep® 70 mL capsule (see fig. 1a) in flow-through applications as the membrane type and bed height are unchanged. Membrane Chromatography The 75 and the 150 mL membrane bed volumes are integrated into housings of the same size. The bed height of the 75 mL capsule is half that of the 150 mL version (Fig. 2). Comparison of a standard capsule originally derived from filter housings without optimization to a void-volume optimized 1 L capsule shows that the dynamic binding capacity at 10% breakthrough can be increased by almost 50%. The difference in void volume is 2.4 liters (–63%); 3.8 L (non-optimized) versus 1.4 L (optimized). The binding capacity at 10% breakthrough is improved from 18.2 mg/mL (non-optimized) to 26.9 mg/L in the optimized version (Fig. 4). The major difference between void-volume-optimized and non-optimized construction is displayed in figure 3. The dynamic binding capacity at 10% breakthrough is increased by 17% for the 75 mL capsule compared to the non-optimized 70 mL (Fig. 5).
