Introduction
The Organelle Lights™ reagent contains a baculovirus that, upon entry into mammalian cells,
directs the expression of autofluorescent proteins that are localized to specific subcellular
compartments and organelles via a signal peptide or protein fusions (Table 2).
Baculoviruses are insect cell viruses. The use of baculovirus for protein production in insect
cells has been used extensively for over two decades; however its application to mammalian
cells is relatively new. The utilization of baculovirus to deliver genes to mammalian cells is
referred to as BacMam technology, and has been recently reviewed.1,2
The BacMam technology has several key features: 1) high transduction efficiency across
a broad range of cell types, including primary, stem, and neuronal cells, 2) minimal
microscopically observable cytopathic effects, 3) highly reproducible and titratable expression,
4) biosafety level 1 reagent as it will not replicate in mammalian cells, and 5) simultaneous
delivery of multiple genes. Overall, the BacMam technology is a straightforward method of
gene delivery with few or no observable side effects.
The Organelle Lights™ reagent combines the content of a targeted fluorescent protein with the
viral delivery of the BacMam technology for simple, specific, and robust protein expression in
mammalian cells.
The Organelle Lights™ reagent allows visualization of specific parts of living cells using
fluorescence microscopy, flow cytometry, or high content imaging (HCI). Each kit contains a
virus solution with a targeting sequence fused to a fluorescent protein gene sequence already
incorporated into the viral genome and an enhancer solution for increased expression of the
chimera. If the potential effects of baculovirus on your cell type are a concern, use Cellular
Lights™ Null (Control) reagent that lacks any mammalian genetic elements. The null (control)
reagent allows determination of potential baculovirus-mediated effects and background
fluorescence.

Note: Cellular Lights™ and Organelle Lights™ reagents utilize the first generation of BacMam
delivery technology, BacMam 1.0. Recent improvements to this system, including the addition
elements that greatly enhance transduction efficiency and expression levels, has warranted
the conversion of these reagents to the next generation products, CellLight™ BacMam 2.0.
Conversion of the Cellular Lights™ and Organelle Lights™ BacMam 1.0 to CellLight™ BacMam
2.0 is expected to be completed by the end of 2010. Refer to www.invitrogen.com for current

Materials Required but Not
Provided
Dulbecco’s Phosphate Buffered Saline (D-PBS) without Ca2+ or Mg2+
Cell types successfully transduced using BacMam delivery technology include: HeLa, CHO
M1WT3, CHO K1, COS-7, SKBR3, HepG2, U-2OS, NIH 3T3, HEK 293, HUVEC, rat
primary astrocytes, LAA progenitor cells (adult stem cells), primary cardiac muscle cells,
primary skeletal muscle cells, NT2 human embryonal carcinoma cells, human mesenchymal
stem cells, porcine cardiac stem cells, primary human arterial smooth muscle cells, primary
human lung fibroblasts, rat neuronal stem cells, and spiral ganglial neurons. Currently, the
delivery system does not work well for hematopoietic or macrophage cells. For a complete list
of cells compatible for use with Cellular Lights™ and Organelle Lights™, refer to our website
(www.invitrogen.com).
Before You Begin
1.1 To use the Organelle Lights™ transduction reagents, plate ~1 × 106–4 × 106 mammalian cells
per well in a 100-mm dish. Allow cells to adhere and grow for approximately 4–24 hours at
37°C, 5% CO2 before proceeding with the transduction (see Figure 1 for an overview of the
transduction process). Optimal transduction occurs when cells are 70–80% confluent.
1.2 Prepare the BacMam enhancer solution. Reconstitute the entire vial of enhancer
(Component B) in 25 µL DMSO (Component C) for a 1,000X solution.
Note: The enhancer solution is stable for multiple freeze/thaw cycles under the recommended
storage conditions. If desired, you may divide the solution into aliquots following reconstitution.
Experimental Protocols
Transduction with Organelle
Lights™ Reagent
If desired, you may try a range of reagent concentrations around the dilution given below
(see step 2.1) in your initial transduction experiments with a particular cell type to determine
the optimal amount of reagent to use. For all colors of Organelle Lights™ reagents, too little
virus will result in transduction of only a few cells. The novel TagRFP is a monomeric red
fluorescent protein obtained from the sea anemone Entacmaea quadricolor, and is about
three times brighter than mCherry13, making it the brightest available monomeric red
fluorescent protein. The CFP and emerald GFP (emGFP) are mutants of the original Aequoria
victoria GFP.
2.1 Prepare 5.5 mL of Organelle Lights™ transduction solution in Dulbecco’s Phosphate Buffered
Saline (D-PBS) without Ca2+ or Mg2+ by combining 2.0 mL of Organelle Lights™ reagent
(Component A) with 3.5 mL of D-PBS.
If you wish to transduce your cells with multiple Organelle Lights™ reagents at the same time,
substitute the additional volume of the reagents for the D-PBS. For example, for transducing
with two Organelle Lights™ reagents combine 2.0 mL of each Organelle Lights™ reagent with
1.5 mL D-PBS. For transducing with three reagents, combine equal amount of each reagent
(1.8 mL of each).
Note: Always protect the Organelle Lights™ transduction reagent (Component A) from light
and place the vial back at 4°C as soon as possible after each use. Exposure to light over time
will decrease the viral titer. Titration of the Organelle Lights™ reagent and/or enhancer may
lead to improved results.

2.2 Aspirate the cell culture medium from a 100-mm dish containing ~1 × 106–4 × 106 adherent
cells, and add 5.5 mL of the diluted Organelle Lights™ transduction solution to the plate. Incubate
the cells at room temperature (20–25°C) in the dark for 2–4 hours with gentle rocking or
shaking.
Note: Some cell types (i.e., primary and stem cells) are sensitive to lack of calcium and
magnesium, and will begin to detach in their absence. You may shorten the incubation
time to 15–30 minutes with these cell types; however, this may decrease the transduction
efficiency.
2.3 Aspirate Organelle Lights™ transduction solution from the cell culture dish.
2.4 Add appropriate cell culture medium with or without serum plus 1X enhancer (i.e., 10 µL of
enhancer per 10 mL media). Incubate the cells for 2 hours in optimal growth conditions (i.e.,
37°C and 5% CO2).
2.5 Aspirate enhancer solution from the cell culture dish and replace with the appropriate cell
culture medium. Incubate cells at 37°C, 5% CO2 for >16 hours to allow for the expression of
the Organelle Lights™ fluorescent protein.
2.6 After incubation, you may perform imaging on live or fixed cells.
2.7 Optional: At this stage, you may trypsinize the cells, freeze in Recovery™ Cell Culture
Freezing Medium, and store in liquid nitrogen for future use.
Imaging the Transduced Cells
3.1 Image cells expressing Organelle Lights™ fluorescent proteins using a fluorescence
microscope or other suitable fluorescence-imaging instrument. Approximate fluorescence
excitation and emission wavelengths in nanometers are: 435/485 for CFP, 485/520 for GFP,
and 555/584 nm for RFP. You may use a FITC filter for CFP and GFP, and a Cy3™ or TRITC
filter for RFP. Refer to Figure 2 for excitation and emission spectra of GFP, CFP, and RFP.
Note: You may observe fluorescence from Organelle Lights™ proteins 16–24 hours following
transduction, which generally persists for more than 5–7 days. Some variation in expression
level from cell to cell within the same culture is normal.

3.2a Optional: You may fix your cells with paraformaldehyde as Organelle Lights™ fluorescence
is well-retained after fixation. To fix, treat cells with 4% paraformaldehyde solution in PBS
for 10–30 minutes at room temperature. If desired, you may permeabilize the cells following
fixation with 0.2% Triton® X-100 in PBS for 5 minutes at room temperature.
Note: Do not use methanol. The fluorescence may be lost. However, if methanol fixation is
necessary for downstream processing, you may detect CFP and GFP proteins using anti-GFP
antibodies and RFP using the anti-RFP antibody, available separately from Invitrogen.
3.2b Following fixation, wash cells by performing two sequential 5 minute washes in PBS. You may
image the cells in PBS, or, for increased photostability, place the cells in ProLong® Gold antifade reagent for imaging.
