HERAEUS®® CYTO SYSTEM FOR THE PREPARATION OF MICROSCOPE SLIDES For the sedimentation of cells and similar biological material directly from the sample to the microscope slide, Heraeus offers the specially developed cyto system. Its design is such that it can be used with all Heraeus general purpose centrifuges. The principle Open cyto containers are placed on to the slide and are loaded with the sample. Due to the centrifugal force the cells contained in the suspension settle on the slide and stick to it. The containers are taken off after centrifugation and the slides are gathered. The cell sediment on the slides is prepared for microscopy through fixing and dying, as usual. ■ Ideal for the production of slide preparations with any cells ■ Suitable for every Heraeus general purpose centrifuge ■ Filter or decanter method ■ Sample volumes of 50 µl to 3 ml are possible ■ Double samples on every slide The accessories The rectangular cyto containers are made of plastic and have two holes, each fully drilled, which are sealed by o-rings on the slide. The tops are sealed with a lid. The two chambers ensure that the sample itself and a reference sample can be prepared on the slide at the same time. The containers are available with cylindrical (1.5 ml) and conical (3 ml) holes. The conical container with its smaller sedimentation area and larger volume, allows cells to be concentrated. The holder, the slides and the container clip together for loading into the centrifuge. The cyto containers are transparent, resistant to chemicals and are autoclavable. The cyto holder is made of a flat metal plate with DIN-slide dimensions (25 x 76 mm). Two side spring metal sheets hold the container on to the slide. Cyto accessories are used for the centrifugation of cells on microscopic slides. The process To achieve high quality results the sample's cell concentration and the sample volume to be used, must be determined. A cell density of 2,000 to 5,000 per mm2 at a diameter of 5 to 10 µm serves as a standard value. The choice of cyto container with their different sedimentation areas is determined by the cell concentration, and volume of the sample. In principle, two methods are possible – the filter method and the decanter method. The filter method A slide is put into the vessel holder and a suitable filter card is positioned between the container and the slide. Micro tubes with a hole in the bottom, which are placed into the container holes, are used to hold cell suspensions. Due to capillary forces, the sample only enters the container once the centrifugal force is applied. The suspended cells reach the glass surface and stick to it. The micro tubes prevent sample loss by stopping a flow on to the filter paper prior to centrifugation. Depending on the suspension's cell concentration, the sample quantity needs to vary. For this method the recommended sample volumes are between 50 to 200 µm. The smaller the sample quantity the better it is absorbed by the filter paper. With larger volumes liquids collect in the bucket's cavity underneath the slide. above which is a little residual liquid, which must be dried prior to dying or fixing. With the filter method the cell sediment is considerably drier than with the decanter method. The decanter method The principle behind this method is that the cyto container and the slide form a tight vessel. The rubber pad which comes with the vessel holder is placed underneath the slide. With the assistance of a pipette the sample material is filled directly into the containers with the o-rings, facing the glass. Following centrifugation the cyto holder with the container is taken out and the liquid is decanted or sucked away. The cell precipitate forms a round spot, The centrifugation For centrifugation the loaded vessel holders are either placed into the Labofuge 400's special cyto bucket, for which an aerosol-tight lid is available, or into the support inserts. Using the support inserts they can be used in any rectangular swing-out bucket. For gentle cell treatment centrifugation needs to take place with the brake switched off. The recommended centrifugation period is five to ten minutes. Depending on the particular sample material, it is essential that all parameters are optimised. WHICH ACCESSORIES FOR WHICH METHOD?
