Arrives!! We are ready to perform
DNA Electrophoresis
Students pour their own agarose gels.
Beginning the DNA
Digest:
1. Add samples of DNA to
micro-test tubes
2. Add the Restriction
Enzyme that will locate
palindromes (patterns) in
the DNA samples and cut
our DNA into fragments.
3. Spin the test tubes using
a micro-centrifuge to be
sure our small samples
are mixed well
4. Provide time for the
restriction enzymes to
work.
DNA Digests work best if we use the
Hot Water Bath!
• Place all DNA samples
into special floating
racks and incubate for
20 minutes. This gives
the restriction
enzyme time to digest
each DNA sample into
fragments.
Now load the DNA samples into the
agarose gels and fill the
electrophoresis chambers.
Place the gels into the Electrophoresis
chambers and turn on the power!

complete the analysis of the suspect DNA
