Cells on Demand™ Custom Services was established to expand this offering and meet each customer’s unique research applications. Expand Your Research Options Modifications to Existing Catalog Products Cells on Demand™ provides the option to manipulate our standard catalog products to suit your experimental needs. Examples include: – – – – – Additional donor characteristics Donor-matched cell sets Pooled donors Cell expansions Additional testing Cell Isolations from Various Tissues Through our extensive tissue sourcing, Cells on Demand™ goes beyond our standard offering to provide cell types of interest from a variety of tissues. Each project adheres to the same ethical sourcing procedures and quality systems of Clonetics™ and Poietics™ Primary Cell Systems. We can provide cell isolations from a variety of species including (but not limited to) human, primate, porcine, canine, feline and murine tissue. We have successfully isolated many cell types, including, but not limited to: – – – – – 2 Human retinal pericytes Human renal podocytes Human renal pericytes Cynomolgus monkey lung microvascular endothelial cells Feline and Porcine renal cells BioResearch – Lonza Cells on Demand™ Custom Services Podocytes at P7, podocin overlay (10x). RAFT™ 3D Culture Services Cell Characterization Lonza’s patented RAFT™ 3D Culture System is designed to closely mimic the in vivo environment of cells within 96-well, or 24-well plate formats. Utilizing Lonza’s RAFT™ System, we can establish 3D Cell Culture Systems tailored to your needs, such as: The Cells on Demand™ Team has a broad experience in many areas of cell biology. Harness this experience and let us develop a cell-based assay for your functional testing of recombinant proteins and small molecules. This can be performed on the recombinant cell lines we have generated, or a specific primary cell type in either 2D or 3D Cell Culture Systems. We offer: – Develop RAFT™ System protocols for your specific cell type – Generate co-cultures and barrier models Ratio Firefly / Renilla – Development of custom cell-based assays with assay optimization and SOP generation 1,000 800 IC50 = 0.04627 μg/mL R2 = 0.9931 600 400 200 Dermal fibroblast fixed and stained after 11 days in RAFT™ System. 0 Generation of Recombinant Cells Cells on Demand™ offers fast and convenient creation of recombinant cell lines. Due to our focus and extensive experience using difficult and hard to transfect cell lines, we have successfully generated single-cell derived clones of cell lines such as sCHO, J774.A1, Jurkat, HL-60, THP1, U937, K562, BJAB, MCF-7, SHSY-5Y and SW480. We can generate specific cell lines overexpressing your gene of interest also including: 1,000 1,000 800 800 600 600 400 400 200 200 0 Clone 24 GFP Clone 28 Clone 34 Clone 39 Clone 51 RLU Mean Fluorescence Intensity – Stably transfected pools and single-cell clone formats (cryopreserved) – Low, medium, and/or high level expressing cell lines including functional characterization in accordance to your requirements – Data sheets including expression characterization and CoA -3 -2 -1 0 1 2 Log Conc. Compound [μg/mL] Compound treatment of sarcoma cells stably expressing an inducible luciferase reporter construct. Cell Line Expansion and Banking Use Lonza as an extension of your cell culture facility. Lonza´s Cells on Demand™ Team can expand your cell line according to specific standards and cryopreserve the cells in your preferred format. This will allow you to focus on your assays and results. We can provide: – Expansions of your specific single-cell derived clone or cell line up to 10e10 – Generation of master cell banks – Generation of working cell banks – QA and delivery of CoA 0 Luciferase Jurkat E6-1 cells were stably transfected with GFP and luciferase reporter constructs. Selected Jurkat E6-1 single-cell derived clones showed diverse co-expression levels of GFP and luciferase. Lonza Cells on Demand™ Custom Services – BioResearch 3 www.lonza.com/research www.lonza.com/cellsondemand Immortalization Services Modify your primary cell to have cell line proliferative capacity. Cell lines are a common screening model because they are easily bulked up to large, homogenous quantities. They often, however, lack the relevant phenotype of the primary cell. In contrast, senescence of primary cells can cause undue frustration for researchers. We overcome the limited lifespan of primary cells with cell immortalization. Lonza has developed a catalog of Clonetics™ Conditionally Immortalized Cells. If your cell type of interest is not in our offering, the Cells on Demand™ Team can produce an immortalized cell line to your requirements. Our offering includes stable immortalization and conditional immortalization. While a stably immortalized cell will regain its proliferative capacity, a conditionally immortalized cell can be switched back to the status of a primary cell after proliferation. We understand that developing these cell lines is not without technical risk, so we have implemented a risk-sharing, milestone approach. On completion of the project we provide a biologically relevant, quality tested screening model.
