Our In-Fusion PCR Cloning Kits are universal, allowing cloning into any vector that has been linearized, provided that your PCR
primers include 15 bp of homology to the ends of the linear vector. To simplify this process, Clontech has created an online primer
design tool. To use our Primer Design Tool, simply provide your vector sequence, partial or complete, and the restriction endonuclease(s)
you will use to linearize the vector. This information is sufficient to determine which 15 bp homology region you must incorporate
into your PCR primers. As an added feature, enter the sequence required to PCR-amplify your insert, and the tool will provide two
complete primer sequences with the required 15 bp of homology incorporated.
Optimized In-Fusion PCR Cloning is achieved through combining a molar rather than a mass ratio of vector to insert (typically a ratio
of 1:2, respectively, is recommended). To further assist in determining the amount of vector and insert to combine in an In-Fusion
cloning reaction, a Molar Ratio Calculator is also provided online. Simply enter the lengths of the vector and the PCR insert, and let
the Calculator do the work for you. Answers will be provided in terms of total nanograms of vector and insert required for any cloning
reaction.
