pastthepolicebulletinstages.Howeverthosecompositesthatassistintheidentificationofasuspect
requireanumberofadditionalinvestigativestepstoprovetheguiltofasuspect.Thelecturewilltalkabout

fingerprints,videoandwitnessidentification.Iwillalsodiscusswhycertainpiecesofevidencearestronger
thanothersandwhymultiplepiecesofevidencetogethermakeforastrongercourtcase.Thispresentation
isfortheforensicartistwhodoesnothaveaninvestigativebackgroundandhasalwayswonderedwhat
occurswiththeircompositeafteritisgiventotheinvestigatorbutisopentoallmembersoftheIAIwho
arecurioushowcompositesworkwiththeirdiscipline.
Thislecturewilldiscussaforensicfacialreconstructioncompletedbythepresenterin2017.Thevictim's
skull,lodgedincement,waslocatedbyacyclistonthesideofamainhighwayinNewYorkstate.Whenthe
skullwasremovedfromthecementbythemedicalexaminer,animpressionofthevictim'sfacewas

swabs,enablingnon-technicaluserstoobtainDNAprofilesrapidlyandwithoutlaboratoryequipment.The
originalANDE™BioChipSet™(BCS)withPowerPlex®16chemistrywasNationalDNAIndexSystem(NDIS)
approvedin2016.Furtherdevelopmentstothesystemhaveallowedanexpansionofthenumberofloci,a
reductionofthequantityofDNArequiredtoobtainaprofileusingaLowDNAContentBCS,andan
Olson
expansionofthesampletypesthatcanbesuccessfullyprocessed.TheDefenseForensicScienceCenterhas

DNAexaminersatDFSCreceivevarioustypesofbiologicalevidence,sometimesinverylargequantities.
Thisrequirestheuseofscreeningmethodstotestforthepresenceofbiologicalfluidsandallowsthe
examinertomakeefficientdecisionsregardingwhatevidenceshouldmoveforwardforDNAtesting.The
currentscreeningmethodsforsemenandsalivautilizedbyDFSCaresemi-quantitative,introducing
potentialfalsepositivesandnotallowingforacorrelationbetweenbodyfluidpresenceandtheabilityto
Meyer
obtainaDNAprofile.Inanattempttoreducefalsepositivesandtodeterminethiscorrelation,a
quantitativemethodforscreeningbothsemenandsalivawasevaluated.Theserologyinstrumentisan
enhancedscreeningtoolthatprovidesquantitativevaluesforbothPSA(semen)andamylase(saliva).Itwas
evaluatedinfourphases:testingitssensitivity,specificity,assessingitsabilitytohandlemixedsamples,and
determiningthecorrelationbetweenthequantvalueoftheinstrumentandtheabilitytoobtainaDNA
profile.Theresultsofthisstudywillbediscussedindetailduringthispresentation.
Themajorityofmycourtroomappearancesthroughoutmycareerhavebeenfortheprosecution.However,
muchofmyprivatecaseworktodayisattherequestofthedefense.InthispresentationI'llbesharingboth

goodandbadaspectsofsomedefensecasesI'veworkedoverthelastthreeyears.Mostoftheissues
involvedocumentation-orthelackthereof-andwilloffersomeguidancetoensureyourcaseworkwill
withstandscrutinyovertime.
Apotentialapplicationofmicrobialgeneticsinforensicscienceisdetectionoftransferofthepubichair
microbiomebetweenindividualsduringsexualintercourseusinghigh-throughputsequencing.Inaddition
totheprimaryneedtoshowwhetherthepubichairmicrobiomeisindividualizing,oneaspectthatmustbe
addressedbeforeusingthemicrobiomeincriminalcaseworkinvolvestheimpactofstorageonthe
microbiomeofsamplesrecoveredforforensictesting.Totesttheeffectsofshort-termstorage,pubichair
sampleswerecollectedfromvolunteersandstoredatroomtemperature(~20oC),refrigerated(4oC),and
frozen(-20oC)for1week,2weeks,4weeks,and6weeksalongwithabaselinesamplefollowedby
amplificationandsequencingoftheV3/V4regionofthe16SrRNAgene.Individualmicrobialprofiles(R2=
0.69)andgender(R2=0.17)werethegreatestsourcesofvariationbetweensamples.Becauseofthis
variation,individualandgendercouldbepredictedusingRandomForestssupervisedclassificationinthis Williams
samplesetwithanoverallerrorrateof2.7%+5.8%and1.7%+5.2%,respectively.Therewasno
statisticallysignificantdifferenceattributabletotimeofsamplingortemperatureofstoragewithin
individuals.Furtherworkonlargersamplesetswillquantifythetemporalconsistencyofindividualprofiles
anddefinewhethertransferbetweensexualpartnerscanbedetected.Forshort-termstorage(<6weeks),
themicrobiomerecoveredwasnotsignificantlyaffectedbythestoragetimeortemperature,suggesting
thatinvestigatorsandcrimelaboratoriescanuseexistingevidencestoragemethods.Disclaimer:The

Thethresholdforsufficiencyinforensiccomparativescienceexaminationsisdifficulttodefine.The
examinationprocess,uniqueandrepeatablefeaturesinsources,levelsofdetailswithinimperfectimages

GraphandtheQualityQuantityCurve,andwherethepotentialuseofexpandingrangesofconclusions
mighttakeuswillbediscussedbythepresenter.
Theneedforinternationalcollaborationinforensiccaseworktosharebestpracticeiswellrecognised.In
forensiceducation,thiscollaborationisstillinitsinfancy.ThispresentationwilloutlinealargeEUfunded
Gwinnett
projectwhichisdevelopingaforensiceducationnetworkacrossmultiplecountriestofacilitate
collaborativeresearchandprovideinternationalprogrammestocreateeffectivetransnational
investigators.ThiswillalsoincludethenextphaseofthisprojectwhichaimstoincludetheUSA.
Duetoinadequaciesinhistoricalhairexaminations,theuseofmicroscopichairevidencehasreduced
severely.Thismeansthatimportantquestionssuchas‘what’,‘where’,‘when’and‘how’areoverlooked.
Thispresentationwilldiscussthefindingsofasurveycompletedbyhairexaminers/researchers.Withinthis Wilkinson
survey,participantsareaskedabouttheevidentialuse,analyticalandinterpretationmethodsof
microscopichairevidence.Thefindingsofthissurveywillthenbeusedtoinformnewmethods.
Forgeryasacommontypeofquestioneddocumentcanbemadebyavarietyofmethods.Thisresearch
studiesthedistributionregularityoftheinkdropletsfromcontinuousinkjetprinteddocumentprintedby
commonbrandinkjetprinterstodeterminewhetheranamountofinkjetprintedpageswereprintedduring Huang
thesameperiodandprinter.Althoughtherearemanyfactorsinfluencetheresult,thisresearch
demonstratedthatmostsamplescouldbeidentifiedproperly.
Wheninvestigatorsareprocessingasceneoritemsinthelabforbiologicalevidence,choosingthemost
effectiveandappropriatecollectionmethodisessential.TheimportanceofDNAevidenceinsolvingboth
activeandcoldcasesiscontinuallyincreasing,andmuchofsocietyexpectsphysicalevidencesuchasDNA
profilestosupportanyprosecution.Wet-vacuumforensicDNAcollectionisbecomingthemethodtoturn Bradley
towhentraditionalmethodsfail.ThisnewmethodenablesinvestigatorstobettercollectDNAmaterial,
especiallyoffofroughand/orporoussurfaces,andoftenleadstostrongerDNAprofilesandmorecases
beingsolved.
TheATFNationalIntegratedBallisticInformationNetwork(NIBIN)isapowerfulleadgeneratingand
investigativetoolforthecrimelaboratoryandlawenforcementagencies.However,oftentimesthe

databaseisnotutilizedtoitsfullestpotential.SincereceivingitsfirstNIBINsystemtwoyearsago,the
presenter'sdepartmentasdevelopedateamworkapproachthathasbeensuccessfulinsolvinggun
violencecrimes.ExamplesandsuggestionsforeffectiveuseoftheNIBINsystemwillbepresented.
InitialstudieshavefoundthatNGShasthepotentialtoreplacecurrentmethodsinhumanidentification
andprojectthattheseNGStechniqueswillbeappliedinforensiclaboratoriesinthenearfuture.TheOffice
oftheChiefScientist(OCS)withintheDefenseForensicScienceCenterisleadingtheefforttobeoneofthe
firstlabsinthenationtoapplythisstate-of-the-arttechnologyinDNAanalysistoForensics.Toadvancethe

applicationofNGSinDNAcasework,OCShaspartneredwithleadersinindustry,joinedasmallgroupof
