Immediately on receipt of your Amoeba culture, remove the cap
from the bottle and place the bottle in a dark cupboard..
MICROSCOPIC EXAMINATION
The Amoeba culture you have received contains 20-25 live amoeba,
a number of chilomonas (a tiny protozoa). The Amoeba are visible
to the naked eye as tiny white specks in the solution when held
against a black background. Allow the culture to stand for several
minutes, then sample with a Pasteur pipette from the bottom of the
culture. Place a drop of culture on a slide and examine under low
power (40x) looking for darkly granular, organisms. Close the iris
diaphragm of the microscope almost entirely to maximise contrast. It
may be necessary to examine several drops to locate the amoeba.
To view mobility gently add a coverslip, relocate and centre under
low power, swing to high power and examine for 5-10 minutes.
Initially, the Amoeba may appear to be dead as their movement is
extremely slow. However if they are observed for several minutes
under the microscope they will be seen to change shape, and
protoplasmic streaming will become visible. Mobility increases with
temperatures up to approximately 25C. Mobility is accelerated if a
microscope with a built in electric light source is used.
CULTURE MEDIUM
It is difficult to culture Amoeba successfully, since they are
sensitive to a range of environmental factors. The following method
can be used to culture your own supply of Amoeba.
Pour Amoeba culture medium (CM 1) into a clean glass petri dish
(rinsed with distilled water) until petri dish is half full. Add 3-4
uncooked rice grains, together with your Southern Biological live
Amoeba culture. Place the lid on the petri dish and allow to stand in
a cool, dark cupboard (approximately 20C) for at least 6-10 weeks.
If your culturing procedure is successful, growth should be apparent
in the form of 100-200 Amoeba approximately 6-10 weeks from the
time of inoculation with the parent culture. However, we should also
mention that only approximately 30% of our own subculturing
dishes yield growth of Amoeba at the end of the 10 week period.
