Laboratory Report Purpose: The purpose of the environmental effects on mitosis lab is to explore biotic and abiotic factors that may affect the rate of mitosis in plants. Hypothesis: In this lab I think we will see that the controlled root tip will have a higher rate of mitosis. Materials: ● hydrochloric acid ● HCI ● stain ● onion root tips in controlled treatment ● Onion root tips in IAA treatment ● Carbol fuchsin solution ● Deionized water ● Cover slips ● Glass slide ● Compound microscope ● Scissors ● Spot plate Procedures: 1. fill the spot plate will all the substances ( deionized water, HCI, stain) 2. Cut your controlled and IAA root tips 3. Put each one into the HCI for 5 minutes 4. Take them out and put them into the carbol fuchsin for 2 minutes 5. Take them out and put them into deionized water for 1 minute 6. Take them out and put them in the stain for 23 minutes 7. Take them out and put them into the deionized water again for 1 minute 8. Remove the root tips from the deionized water and place them on a glass slide and crush the root 9. Place a drop of water on top of the root tip, next place the cover slide on top of the root tip, clean off any excess water 10. Place into the compound microscope and using the the 40x lense observe the root tip Results and Data: When I was looking at my root tip I couldn't see a clear picture of the cells going through mitosis so I got two pictures off the internet in order to be able to complete the lab.The Root growth root had 35 cells in total. It had 14 in interphase and 21 in mitosis. When calculating the chi square value I got 0 for the Root growth onion root. The controlled root had 57 cells in total. It had 33 in interphase and 25 in mitosis. When calculating the chi square value for the controlled onion root tip I got 17.8. So when I combined both answers the total was 17.8. When checking the table it said that if the calculated chisquare value is greater than the critical value obtained from the table for the degrees of freedom and the selected probability value, then the null hypothesis is rejected. This means that the observed difference in the rate of mitosis between the treated sample and the control sample is considered statistically significant.
