Aqueous buffer containing 0.2 M phosphate buffer with
27 mM KCl, 1.37 M NaCl and 0.5% Surfactant P20 (Tween 20).
Sterile filtered. Will yield pH 7.4 when diluted and
supplemented with 2% DMSO.

Storage:
Store at room temperature (precipitation may occur at low
temperature).
The use-before date stated on the bottle label applies to
unopened bottle.

Intended use
PBS-P+ buffer, 10x is used for the preparation of running buffer with
DMSO, which is commonly used for small molecule assays in Biacore
instruments. The 10x buffer is optimized for 2% DMSO, but other DMSO
concentrations may also be used, see “PBS-P+ buffer with other DMSO
concentrations” on page 2.
Prepare PBS-P+ buffer with 2% DMSO
To prepare PBS-P+ buffer with 2% DMSO, do the following:
1 Prepare a 1.02x PBS-P+ solution as follows: Dilute the 10x
solution with deionized, 0.2 µm filtered water, e.g. by mixing
102 ml 10x solution and 898 ml water. This solution is used for
the preparation of running buffer (as described below), solvent
correction stock solutions and samples.
2 Prepare PBS-P+ running buffer with 2% DMSO as follows: Dilute
100% DMSO with 1.02X PBS-P+ solution, e.g. by mixing 20 ml
100% DMSO and 980 ml 1.02X PBS-P+ solution. Refer to
www.gelifesciences.com/biacore for laboratory guidelines for
buffer and sample preparation for direct binding assays in
DMSO.
3 Degas the buffer before use in systems without degasser.
Follow the instructions in the instrument manual for start-up with
new buffer.
PBS-P+ buffer with other DMSO concentrations
If buffer with other DMSO concentrations than 2% is prepared, the
pH-value will change according to Fig 1 (data obtained at room
temperature).

If using PBS-P+ running buffer with additives containing
metal ions, make sure that it is compatible with BIAdesorb
solution 1 before running the Desorb or Desorb and
Sanitize procedures (SDS will precipitate with some
common metal ions such as K+ and Mg2+).
