In case of meat, the specification of the animal species has to be disclosed on the label. In Germany the necessity of identifying meat components to prevent potential misleading of consumers is based on §11 of the German Food and Feed Law. If material from animal species, other than the declared ones, are used as components, or if components are used, whose absence is stated in the labeling, the respective food should be rejected. Tab. 1 specificity results of the ANIMAL ID Horse & Donkey IAAC real-time PCR detection method (examples) In order to enable the authorized food control agencies or quality assurance of the food industry to supervise compliance with the labeling requirements suitable detection methods must be available. Method ® After DNA extraction with SureFood PREP Basic the animal species horse (Equus caballus) and donkey (Equus asinus) will be fast and sure detected with the real-time PCR technology. Horse-DNA is detected in the FAM channel and donkey-DNA is detected in the Cy5 channel. In the VIC/HEX channel it is possible to detect animal DNA in the sample as well as the amplification control in a sample with no animal DNA inside (internal inhibition control). Through the prepared limit of detection it is possible to distinguish intentional admixture from technological unavoidable contaminations. The assay limit of detection depends on sample matrix, processing grade, DNA-preparation and DNAcontent. Donkey detection Specificity For verifying the specificity of ANIMAL ID Horse & Donkey IAAC real-time PCR, DNA of reference material from BfEL (former Bundesforschungsanstalt für Ernährung und Lebensmittel) and potentially horse and donkey free food components were analysed. For the horse and donkey real-time PCR detection method no cross reactivity could be detected (see table 1).
