To learn to see cell parts more easily by using cell staining techniques. 2. Methods and Materials: a. Plant Cells (Elodea Plant) 1. Get a leaflet (the thinnest part of the leaf tip that allows more light up through the specimen) from your teacher. Place it on a slide, cover with a cover slip. 2. Search for the cells using low power (4x), sketch what you see. 3. Look at the cells again using med (10x) & high (40x), sketch and label your drawings. 4. Use color and details in every sketch. b. Animal Cells (your own cheek cells) 1. Place one drop of Methylene Blue STAIN on your slide. 2. Get a toothpick from your teacher. GENTLY run the rounded edge of the toothpick along the inside of your cheek as demonstrated. 3. Rub it around in the drop of stain (releasing the cheek cells). 4. Cover with a cover slip and throw away the toothpick! 5. Search for the cells using low power (4x), sketch what you see. 6. Adjust the diaphragm to #3 instead of bright light if necessary. 7. Look at the cells again using med (10x) & high (40x), sketch and label your drawings. 8. Use color and details in every sketch. 3. Hypothesis: not needed 4. Observations and Sketches: ok to use the back of your sheet for your lab report if it’s neatly drawn, in color and labeled (at least 3 organelles on 10x and 40x sketches, magnification on all). 5. Conclusion: How are plant and animal cells the same and how are they different? (don’t include cell staining information). Do the extra credit first to help you with the conclusion. 6. Analysis Questions: 1. Why did we use Methylene Blue Stain on the human cheek cells and not on the Elodea cells? 2. Name the 2 parts of the cell that you saw in the plant cell that you did not see in the animal cell. 3. How do you think the different jobs (functions) or the cells affect their shape (structure)?
