Material Number: Alternate Name: Size: Vol. per Test: Clone: Immunogen: Isotype: Reactivity: Storage Buffer: Description The 90220 monoclonal antibody specifically binds to the beta subunit (IL-10R beta/IL-10RB/IL-10Rβ) of the heteromeric IL-10 Receptor complex. This type I transmembrane glycoprotein is also known as, subunit 2 (IL-10R2) or CD210b. The IL-10 Receptor complex is formed by the binding of dimeric IL-10 to two cell surface alpha subunits of the IL-10 Receptor (CD210a/IL-10RA/IL-10Rα). This complex can then recruit two CD210b subunits resulting in the transduction of intracellular JAK/STAT pathway signals. CD210b is also a component of cytokine receptor complexes specific for IL-22, IL-26, IL-28 and IL-29. CD210b is expressed by a variety of cell types including T cells, B cells, NK cells, monocytes and macrophages. Multiparameter flow cytometric analysis of IL-10Rβ (CD210b) expression on human peripheral blood leucocytes. Human whole bloo was stained with PE Mouse Anti-Human HLA-DR (Cat. No. 555812/556644/560943) and FITC Mouse Anti-Human CD14 (Cat. No. 555397/557153/561712) antibodies and either Alexa Fluor ® 647 Mouse IgG1 κ Isotype Control (Cat. No. 557714) or Alexa Fluor ® 647 Mouse Anti-Human IL-10Rβ (CD210b) antibody (Cat. No. 564372). Erythrocytes were lysed with BD Pharm Lyse™ Lysing Buffer (Cat. No. 555899). Flow cytometric analysis was performed using a BD™ LSR II Flow Cytometer System. Panel 1. Two-parameter contour plots show the correlated expression of isotype control (Left Plot) or IL-10Rβ (CD210b) (Right Plot) versus side-light scatter (SSC-A) characteristics for gated events with the forward light scatter properties of total viable leucocytes. Panel 2. For gated events with the forward and side-light scatter characteristics of viable mononuclear leucocytes, the two-color contour plots show the correlated expression of CD14 (Left Plot) or IL-10Rβ (CD210b) (Right Plot) versus HLA-DR for HLA-DR+ (and HLA-DR-) lymphocytes and CD14+HLA-DR+ monocytes. Preparation and Storage Store undiluted at 4°C and protected from prolonged exposure to light. Do not freeze. The monoclonal antibody was purified from tissue culture supernatant or ascites by affinity chromatography. The antibody was conjugated to Alexa Fluor® 647 under optimum conditions, and unreacted Alexa Fluor® 647 was removed.
